益气化瘀醒脑汤对急性脑缺血模型大鼠脑组织病理学、神经细胞凋亡及氧化应激损伤的影响

摘  要:目的:探讨益气化瘀醒脑汤对急性脑缺血模型大鼠脑组织病理学、神经细胞凋亡及氧化应激损伤的影响。方法:采用改良闭塞线栓法栓塞大脑中动脉法建立急性脑缺血大鼠模型,60只大鼠随机分为6组,每组10只。造模24 h后正常对照组和模型组给予生理盐水灌胃,益气化瘀醒脑汤低、中、高剂量组给予益气化瘀醒脑汤(5、10、15 g/kg)灌胃,阳性组给予尼莫地平片(10 mg/kg),1次/d,连续给药7 d。末次给药24 h后评价神经功能损伤程度,TTC染色比较大脑梗死面积,HE染色观察大脑病理形态变化,TUNEL法检测神经细胞凋亡,并检测外周血氧化应激损伤指标(SOD、T-AOC、GSH-Px)。结果:与正常组比较,模型组神经功能损伤评分和脑梗死体积显著增加,益气化瘀醒脑汤低、中、高剂量组和阳性组神经功能损伤评分和脑梗死体积显著低于模型组(P<0.05)。与正常对照组比较,模型组梗死灶内细胞和血管坏死、脑组织细胞疏松、形态改变、间质水肿;益气化瘀醒脑汤低、中、高剂量组和阳性组脑组织损伤减轻,细胞稍显疏松,水肿减轻。与正常对照组比较,模型组神经细胞大量凋亡;与模型组比较,益气化瘀醒脑汤低、中、高剂量组和阳性组神经细胞凋亡显著降低(P<0.05)。与正常对照组比较,模型组血MDA含量升高,SOD、T-AOC和GSH-Px活性降低;与模型组比较,益气化瘀醒脑汤低、中、高剂量组和阳性组MDA含量显著降低,SOD、T-AOC和GSH-Px活性显著升高(P<0.05)。结论:益气化瘀醒脑汤可显著降低急性脑缺血模型大鼠神经功能损伤和脑梗死体积,改善脑组织损伤,减少神经细胞凋亡,其机制可能与减轻氧化应激损伤水平有关。Objective:To investigate the effects of Yiqi Huayu Xingnao Decoction on brain histopathology,neuron apoptosis and oxidative stress injury in rats with acute cerebral ischemia.Methods:The rat model of acute cerebral ischemia was established by occlusion of the middle cerebral artery with modified occlusion line embolization method.60 rats were randomly divided into 6 groups,and 10 rats were in each group.The normal control group and the model group were intragastrically administered with saline for 24 hours,and Yiqi Huayu Xingnao Decoction was administered to the low,middle and high dose groups(5、10、15 g/kg)by intragastric administration,the positive group was given Nimodipine(10 mg/kg),once a day,for 7 d.24 hours after the last administration,the degree of neurological damage was evaluated.TTC staining was used to compare the area of cerebral infarction,HE staining was used to compare the pathological changes of the brain,the apoptosis of nerve cells was detected by TUNEL method,and the oxidative stress injury index in peripheral bloods was detected.Results:Compared with the normal group,the neurological impairment score and cerebral infarction volume of the model group were significantly increased,and the Yiqi Huayu Xingnao Decoction and positive group neurological impairment score and cerebral infarction volume were significantly lower than those of the model group(P<0.05).Compared with the normal group,model group showed that cells and vascular necrosis in the infarcted area,loosening of brain tissue cells,cell morphology change,and interstitial edema.Yiqi Huayu Xingnao Decoction and positive group showed that brain tissue damage was reduced,slightly loosened cells,and edema lighten.Compared with the normal group,there was a large number of apoptosis in the model group,compared with the model group,the apoptosis in the Yiqi Huayu Xingnao Decoction low,middle,high dose group and the positive group were decreased significantly(P<0.05).Compared with the normal group,the MDA in the model group was significantl
