西洋参叶山楂叶有效部位组合物活血化瘀作用的实验研究

摘  要:目的:研究西洋参叶与山楂叶有效部位(XS)组合物对以皮下注射肾上腺素及冰浴刺激致大鼠急性血瘀模型血液流变学和正常大鼠血小板聚集率的影响。方法:①将大鼠按体质量随机分为正常对照组、急性血瘀模型组,益心酮分散片组,XS组合物高、中、低(58.8、29.4、14.mg/kg)剂量组。对照组和模型组灌胃同体积的纯净水,其余各组灌胃相对应的药物,连续7 d,除正常对照组大鼠外,其余各组大鼠采用皮下注射肾上腺素后进行冰水浸泡制作大鼠急性血瘀模型。末次灌胃给药1 h后,将大鼠麻醉后腹主动脉取血,测定血液流变学指标。②将大鼠随机分为空白对照组,益心酮分散片组,XS组合物高、中、低剂量组,空白对照组灌胃同体积的纯净水,其余各组灌胃相对应的药物,连续7 d,末次给药1 h后,于大鼠麻醉后腹主动脉取血,测大鼠血小板聚集率。结果:①与正常对照组比较,模型组大鼠全血黏度、血浆黏度、红细胞压积、血沉明显升高(P<0.01),大鼠急性血瘀模型复制成功。与模型组比较,XS组合物高、中剂量能明显降低急性血瘀模型大鼠的全血黏度(P<0.01,P<0.05)。②XS组合物高、中剂量组与空白对照组比较均能明显降低大鼠血小板的1、3、5 min及最大聚集率(P<0.01,P<0.05)。结论:XS组合物可明显改善急性血瘀模型大鼠血液流变学的异常改变,降低血液的高黏状态,抑制血小板聚集率,有活血化瘀作用。Objective:To study the effects of effective parts(XS)compositions of Panax quinquefolium and Hawthorn leaves on hemorheology in the models of acute blood stasis induced by subcutaneous injection of adrenaline and ice bath stimulation and platelet aggregation in normal rats.Methods:①According to body weight,the rats were randomly divided into normal control group,acute blood stasis model group,positive group,XS compositions of high,medium and low dose(58.8,29.4,14.7 mg/kg)groups.The normal control and model groups were given the same volume of pure water,and the other groups were given corresponding drugs,for 7 days.Except for the normal group,rats in other groups were injected subcutaneously with epinephrine and immersed in ice water to prepare acute blood stasis model.One hour after the last intragastric administration,blood was taken from the abdominal aorta of anesthetized rats to measure the hemorheological indexes.②The rats were randomly divided into blank control group,positive group,XS compositions of high,medium and low dose groups.Blank control group was given the same volume of pure water,and other groups were given corresponding drugs for 7 days.After one hour of the last administration,blood was taken from abdominal aorta after anesthesia to measure the platelet aggregation rate of the rats.Results:①Compared with normal control group,whole blood viscosity,plasma viscosity,hematocrit and erythrocyte sedimentation rate of rats were significantly increased in model group(P<0.01),and acute blood stasis model was successfully replicated.Compared with model group,high and medium doses of XS compositions significantly reduced the whole blood viscosity of acute blood stasis model of rats(P<0.01,P<0.05).②Compared with blank control group,XS compositions in high and medium dose groups significantly reduced the platelet aggregation rate of rats at 1,3 and 5 min and the maximum aggregation rate of rats(P<0.01,P<0.05).Conclusion:XS compositions can significantly improve the abnormal changes of hemorheology
