摘  要:探讨慢病毒介导SOX2转染大鼠SCI星形胶质细胞是否转化成双皮质素(doublecortin,DCX)-阳性的神经干细胞或神经元,从而影响SCI后功能恢复.60只成年雄性SD大鼠随机分为5组:Sham组和SCI组(注射生理盐水),SOX2-NC组(注射慢病毒),SOX2-OE组和SOX2-OE+VPA(注射过表达SOX2慢病毒).术后各观察点对各组行BBB评分和Rivlin实验,HE染色观测脊髓形态,甲苯胺蓝染色观测尼氏小体数及形状,免疫组化及WB检测神经元再生情况.实验结果发现术后各组运动评分都降低,随着时间发展评分缓慢上升,SOX2-OE和SOX2-OE+VPA组术后1 w及其后均高于SCI组;HE染色示Sham组脊髓层次清晰;SCI组脊髓灰白质层次不清;SOX2-OE和SOX2-OE+VPA组:脊髓灰白质层次尚清楚;尼氏染色示SOX2-OE、SOX2-OE+VPA组Nissl小体阳性数多于SCI组;免疫组化及Western Blot示:SOX2-OE和SOX2-OE+VPA组DCX-阳性神经干细胞、增殖神经元数多于SCI组(P<0.05).SOX2转录因子转染脊髓损伤局部星形胶质细胞后促进DCX-阳性神经干细胞和神经元增多.本实验为SCI医治提供潜在的治疗手段,具有重要的意义和价值.This paper explores whether the lentivirus-mediated SOX2 transfection of rat SCI astrocytes is transformed into doublecortin(DCX)-positive neural stem cells or neurons,thereby affecting the functional recovery after SCI.60 adult male SD rats were randomly divided into 5 groups:Sham group and SCI group(injection of saline),SOX2-NC group(injection of lentivirus),SOX2-OE group and SOX2-OE+VPA(injection of overexpression SOX2 slow virus).BBB score and Rivlin test were performed at each observation point after operation.The spinal cord morphology was observed by HE staining,the number and shape of Nissl’s body were observed by toluidine blue staining,and the neuron regeneration was detected by immunohistochemistry and WB.The experimental results found that the motor scores of each group decreased after the operation,and the scores slowly increased with time.The SOX2-OE and SOX2-OE+VPA groups were higher than the SCI group at 1 week and after the operation;HE staining showed that the spinal cord level of the Sham group was clear,The gray matter levels of the spinal cord in the SCI group are unclear,SOX2-OE and SOX2-OE+VPA groups:the gray matter levels of the spinal cord are still clear;Nissl staining shows that the number of positive Nissl bodies in the SOX2-OE and SOX2-OE+VPA groups is more than that in the SCI Group;immunohistochemistry and Western Blot showed that the numbers of DCX-positive neural stem cells and proliferating neurons in SOX2-OE and SOX2-OE+VPA groups were more than those in SCI group(P<0.05).The transfection of SOX2 transcription factor into astrocytes in localized spinal cord injury promotes the increase of DCX-positive neural stem cells and neurons.This experiment provides a potential treatment method for SCI,which has important significance and value.
