摘 要:【目的】分析侵染贵州火龙果的仙人掌X病毒(Cactus virus X,CVX)及其NTU株系的基因组序列遗传变异,为建立分子检测和防控体系奠定理论基础。【方法】对感染病毒的样品采用宏转录组测序和生物信息学分析,分离病毒基因组序列并分析分子变异和系统进化。【结果】获得4条CVX(CVX-LW、CVX-RF、CVX-ZNS和CVX-ZYP)和5条CVX-NTU(CVX-NTU-LW、CVX-NTU-RF、CVX-NTU-ZNF、CVX-NTU-ZNS和CVX-NTU-ZYP)贵州火龙果分离物近全长基因组序列,长度为6494~6603 bp,覆盖参考基因组96.2%~99.6%;CVX贵州分离物之间的基因组序列一致性为98.3%~98.6%,与已报道的其他分离物基因组序列一致性为97.1%~98.0%;CVX-NTU贵州分离物之间的基因组序列一致性为98.1%~98.8%,与已报道的其他分离物基因组序列一致性为97.6%~99.1%;CVX和CVX-NTU群体未出现明显的遗传分化。【结论】研究获得的CVX和CVX-NTU贵州火龙果分离物近全长基因组序列表现较高的一致性,未发生明显遗传变异。【Objective】The genetic variation in genome sequences of Cactus virus X(CVX)infecting Hylocereus undatus in Guizhou and their NTU strains is analyzed to lay the theoretical basis for establishing molecular determination and control system.【Method】The samples infecting virus are studied by metatranscriptomic sequencing and bioinformatics analysis,then viral genome sequences are obtained,and the molecular variation and phylogeny are analyzed.【Result】Four near full-length genome sequences of CVX Guizhou H.undatus isolates(CVX-LW,CVX-RF,CVX-ZNS and CVX-ZYP)and five near full-length genome sequences of CVX-NTU Guizhou H.undatus isolates(CVX-NTU-LW,CVX-NTU-RF,CVX-NTU-ZNF,CVX-NTU-ZNS and CVX-NTU-ZYP)are obtained,with length of 6494~6603 bp,covering 96.2%~99.6%of the reference genome.The genome sequence identities among CVX Guizhou H.undatus isolates is 98.3%~98.6%,and 97.1%~98.0%with other reported CVX isolates.The genome sequence identities among CVX-NTU Guizhou H.undatus isolates is 98.1%~98.8%,and 97.6%~99.1%with other reported CVX-NTU isolates.CVX and CVX-NTU population have no obvious genetic differentiation.【Conclusion】The near full-length genome sequences of CVX and CVX-NTU Guizhou H.undatus isolates in the study shows high identity,and has no obvious genetic variation.
