摘 要:【目的】了解烤烟感病苗与健康烟苗根围微生物的群落结构与多样性特征,为指导烟草苗期病害的防控提供参考。【方法】采用高通量测序技术对感病与健康烟苗根围基质进行微生物群落结构与多样性分析。【结果】感病与健康烟苗根围基质优势细菌和真菌微生物群落结构存在共性与差异。感病与健康烟苗根围基质优势细菌门均为变形菌门、拟杆菌门和放线菌门,优势真菌门均为子囊菌门、担子菌门和罗兹菌门;优势细菌属均为未分类的蓝细菌、德沃斯氏菌属、假单胞菌属、鞘脂单胞菌属、寡养单胞菌属、柄杆菌属、罗河杆菌属、Pseudolabrys、噬氢菌属和Reyranella,优势真菌属均为金孢子菌属和假裸囊菌属。感病烟苗基质特有优势细菌属为Dongia、慢生根瘤菌属和纤维菌属,特有优势真菌属为锥毛壳属、螺菌属、丝衣霉属、曲霉属和Pseudeurotium,其中感病样品(JZT2)中亡革菌属含量最高。健康烟苗根围基质特有的优势细菌属为Terrimonas、地杆菌属和Romboutsia,特有优势真菌属为节丛孢属。健康烟苗根围基质真菌和细菌的Alpha多样性指数(Shannon、Chao1、ACE)均高于感病烟苗根围基质。经功能预测,感病与健康烟苗根围基质细菌主要分布于代谢通路;真菌主要分布于未定义腐生菌和植物病原菌这2个生态功能群,其中感病烟苗根围基质的植物病原菌类群较健康烟苗多。【结论】烟苗根围基质微生物群落结构复杂,其类群组成与烟苗感病密切相关。健康烟苗根围基质真菌和细菌的Alpha多样性指数均高于感病烟苗根围基质;感病烟苗根围基质的植物病原菌类群较健康烟苗多。【Objective】The microbial community structure and diversity characteristics in rhizosphere substrate of diseased and healthy tobacco seedling are investigated to provide a reference for the control and prevention of tobacco seedling diseases.【Method】The microbial community structure and diversity in rhizosphere substrate of diseased and healthy tobacco seedling are analyzed by using high-throughput sequencing technology.【Conclusion】There are both similarities and differences in the microbial community structure of dominant bacteria and fungi in the rhizosphere substrate of diseased and healthy tobacco seedling.The dominant phyla of bacteria in rhizosphere substrate of diseased and healthy tobacco seedling are proteobacteria,bacteroidetes and actinobacteria.The dominant fungal phyla are ascomycota,basidiomycota and rozellomycota.The dominant bacterial genera are unidentified cyanobacteria,Devosia,Pseudomonas,Sphingopyxis,Stenotrophomonas,Caulobacter,Rhodanobacter,Pseudolabrys,Hydrogenophaga and Reyranella,and the dominant fungal genera are Chrysosporium and Pseudogymnoascus.The unique dominant bacterial genera in diseased tobacco seedling substrate are Dongia,Bradyrhizobium and Cellulomonas;The unique dominant fungal genera are Coniochaeta,Neobulgaria,Byssochlamys,Aspergillus and Pseudeurotium,and the content of Thanatephorus is the highest in JZT2 sample.The unique dominant bacterial genera in healthy tobacco seedling rhizosphere substrate are Terrimonas,Pedobacter and Romboutsia,and the unique dominant genus is Arthrobotrys.The Alpha diversity index(Shannon,Chao1 and ACE)of fungi and bacteria in healthy tobacco seedling rhizosphere substrate are all higher than those in diseased tobacco seedling substrate.The function prediction shows that the bacteria in diseased and healthy tobacco seedling rhizosphere substrate are mainly distributed in metabolic pathway;The fungi are mainly distributed in the two ecological function groups of undefined saprophytes and plant pathogen,of which the plant pathogen in
