摘  要:目的利用数据挖掘构建组合网络,通过生物信息学分析获取肉桂醛改善糖代谢作用靶点。方法首先应用网络药理学方法挖掘出数据库中肉桂醛改善糖代谢作用的32条通路;再基于R软件包“graphite”将数据库中的通路转换为生物网络,在线进行富集分析;然后用Cytoscape构建代谢组学与网络药理学的组合网络;最后推测改善糖代谢作用的关键靶点和潜在的关联机制。结果筛选出10个与肉桂醛改善糖代谢有关的关键靶点,其中IRS-1、AMPK-1、AMPK-2、PRKAB-1和PRKAB-2这5个靶点最有可能受肉桂醛调控。结论采用网络比较和比对方法建立生物通路组合网络,分析肉桂醛的候选关键靶点的方法是有效可行的。Objective To construct a combinatorial network by data mining and obtain the targets of cinnamaldehyde in improving glucose metabolism through bioinformatics analysis.Methods Firstly,32 pathways of cinnamaldehyde in improving glucose metabolism in the database were excavated by the network pharmacology method.Then the pathways in the database were transformed into biological networks based on R software package"graphite",and enrichment analysis was carried out online.Cytoscape was used to construct a combinatorial network of metabonomics and network pharmacology afterwards.Finally,the key targets and potential correlation mechanisms in improving glucose metabolism were speculated.Results Ten key targets related to improving glucose metabolism by cinnamaldehyde were screened,among which IRS-1,AMPK-1,AMPK-2,PRKAB-1 and PRKAB-2 were most likely regulated by cinnamaldehyde.Conclusion It is effective and feasible to establish a combinatorial network of biological pathways by network comparison and comparative method to analyze the candidate key targets of Cinnamaldehyde.
