摘  要:非热等离子体是新型的肿瘤治疗技术,小分子药物草氨酸钠系乳酸脱氢酶特异性抑制剂,对肿瘤治疗具有潜在价值.笔者采用介质阻挡放电装置(dielectric barrier discharge,简称DBD)为等离子体发生源,通过检测细胞活力,发现等离子体与草氨酸钠联用组细胞活力显著降低;通过检测细胞凋亡发生率、线粒体膜电位变化和凋亡相关蛋白的表达,发现二者联用显著促进细胞凋亡;通过检测AMPK和p-AMPK的表达,发现联用组细胞中AMPK磷酸化显著增强;胞内超氧阴离子水平检测证实,联用组胞内氧化自由基水平显著上升;胞内能量代谢水平检测表明,联用组细胞能量代谢被显著抑制.以上结果提示,等离子体与草氨酸钠联用最终导致黑色素瘤细胞下游凋亡通路过度激活,从而增强了对肿瘤细胞的杀伤作用.该研究证明了草氨酸钠可增强人黑色素瘤细胞的等离子体敏感性,可为将来等离子体肿瘤治疗提供一定的理论基础和指导.Non-thermal plasma(NTP) is a new technique for the cancer therapy.Oxamate,a specific inhibitor of lactate dehydrogenase,has been proved to have potential multi-functions in antitumor.Non-thermal dielectric barrier discharge plasma was employed in the present work.It was found that the combined treatment with oxamate and NTP exposure significantly enhanced the killing effect on melanoma cells with cell viability detection.Results of the apoptosis rate,the cellular MMP(mitochondrial membrane potential) and the expression of apoptosis-related proteins showed the combined treatment distinctly promoted apoptosis.Enhanced phosphorylation of AMPK and production of reactive oxygen species were found after the combined treatment.In addition,analysis of the cellular energy metabolism indicated the distinct reduced energy supply after the combined treatment.These results suggested the enhanced activation of apoptotic pathway in melanoma cells and the killing effect with the treatment of oxamate plus NTP.The authors hope their results can provide some new insights to the tumor plasma treatment in the future.
