摘 要:目的:观察补充吡咯喹啉醌(PQQ)对力竭运动所致小鼠心肌损伤、氧化应激和细胞凋亡的影响,探讨其对运动性心肌损伤产生保护作用的可能机制。方法:40只7周龄雄性昆明小鼠,按体重随机分为正常对照组(NC组)、力竭运动组(E组)、PQQ小剂量(5mg/kg/d)+力竭运动组(LE组)、PQQ中剂量(10mg/kg/d)+力竭运动组(ME组)、PQQ大剂量(20mg/kg/d)+力竭运动组(HE组),每组8只。力竭运动各组小鼠尾部负重3%体重的负荷,进行6d/周,1次/d的力竭游泳运动,共2周。采用Elisa和化学比色法测定小鼠血清和心肌组织生化指标,Real-time PCR法检测心肌凋亡因子Caspase-3、Bax和Bcl-2的基因表达。结果:(1)与NC组相比,E组小鼠血清cTnⅠ水平和CK-MB活性显著升高(P<0.05,P<0.01);心肌组织SOD和GSH-PX活性明显降低(P<0.01,P<0.05);MDA含量、Bax/Bcl-2比值和caspase-3 mRNA表达水平显著增加(P<0.01)。(2)与E组相比,除HE组小鼠血清cTnⅠ水平和心肌GSH-PX活性变化没有显著性外,LE、ME和HE组小鼠,血清cTnⅠ水平和CK-MB活性显著下降(P<0.05 or P<0.01);心肌组织SOD和GSH-PX活性明显升高(P<0.05 or P<0.01),MDA含量、Bax/Bcl-2比值和caspase-3 mRNA表达水平显著下降(P<0.05 or P<0.01)。结论:PQQ可以通过降低心肌细胞的氧化应激和细胞凋亡水平减轻力竭运动引起的心肌损伤。Objective:This study assessed the effects and mechanism of Pyrroloquinoline Quinone(PQQ)on myocardial injury,oxidative stress and apoptosis induced by exhaustive exercise in mice.Methods:Forty 7-week-old male Kunming mice were randomly divided into five groups.Group NC was a sedentary control group,Group E,LE,ME and HE performed exhaustive swimming(6 days/week,1 time/day)。All mice had normal diets,but those mice in Group LE,ME and HE were supplemented with 5,10,20 mg/kg/d PQQ respectively.After 2 weeks,all mice were euthanized,serum and myocardiums were collected,the biochemical parameters were determined by Elisa and chemical colorimetry,and the mRNA expression of myocardial apoptotic factors Caspase-3,Bax and Bcl-2 were detected by Real-time PCR.Results:(1)Compared with group NC,a significant increase was observed in the serum level of cTnⅠand the activity of CK-MB in the group E(P<0.05,P<0.01);The activities of SOD and GSH-PX in the myocardium were significantly lower than group NC(P<0.01,P<0.05),while the level of MDA、Bax/Bcl-2 ratio and the mRNA expression of caspase-3 were significantly higher than group NC(P<0.01).(2)Compared with group E,except the level of serum cTnⅠand the activity of myocardial GSH-PX change is not significant in the group HE,the serum level of cTnⅠand the activity of CK-MB decreased significantly in the group LE,ME and HE(P<0.05 or P<0.01);The activities of SOD and GSH-PX in myocardium were significantly increased(P<0.05 or P<0.01),while the level of MDA、Bax/Bcl-2 ratio and the mRNA expression of caspase-3 were significantly decreased in the group LE,ME and HE(P<0.05 or P<0.01).Conclusion:PQQ can relieve myocardial injury caused by exhaustive exercise through reducing oxidative stress and apoptosis levels of cardiomyocytes.It was found that among the three doses of PQQ intervention.
