摘 要:本研究利用PCR方法扩增新城疫病毒(NDV)rGM株的F基因,并将其克隆至真核表达载体pCAGGS,构建重组表达质粒pCAGGS-F。采用离子交联法将壳聚糖、MgSO4、pCAGGS-F以及三聚磷酸钠(TPP)混合,制备成新城疫DNA纳米颗粒疫苗。IFA试验检测结果表明,F重组蛋白成功表达。制备的纳米颗粒疫苗通过点眼滴鼻免疫2周龄SPF鸡2次后,对10^(3) EID_(50) NDV强毒株rGM保护率可达100%。此试验为新城疫DNA纳米疫苗等的研制奠定基础。In this study,the structural protein F gene of Newcastle disease virus(NDV)strain rGM was amplified by PCR and the PCR products were cloned into eukaryotic expression vector pCAGGS.DNA nanoparticle vaccine was then prepared by mixing of PCAGGS-F,chitosan,sodium tripolyphosphate(TPP)and ion crosslinking.The expression of the F gene was demonstrated in IFA.Twoweek-old SPF chickens were intranasally administered twice with the nanoparticle vaccine preparation and then challenged with 10^(3) EID_(50) NDV virulent strain rGM.As a result,all birds were protected.This experiment provided a promising prospect for the development of NDV DNA nanoparticle vaccine.
