摘 要:为了建立同时检测黄芩药材中黄芩苷、汉黄芩苷、黄芩素、汉黄芩素4种成分的一测多评方法。采用高效液相色谱法进行测定,以黄芩苷为内参物,建立汉黄芩苷、黄芩素、汉黄芩素的相对校正因子与相对保留值,采用不同规格与配置的仪器对试验结果进行验证,以一测多评法检测15批黄芩药材样品并与外标法检测结果进行比较。结果表明,汉黄芩苷、黄芩素、汉黄芩素与内参物黄芩苷间的相对校正因子分别为1.02,1.66,0.93;RSD分别为2.03%,3.25%,1.85%(n=15)。相对保留值分别为2.11,2.80,3.57;RSD分别为0.83%,0.96%,0.98%(n=15)。精密度、稳定性、重复性试验、不同仪器验证试验结果RSD值均小于3.5%(n=6);采用一测多评法与外标法检测15批样品,汉黄芩苷误差为0.53%~0.70%、黄芩素误差为-1.30%~3.33%、汉黄芩素误差为-2.99%~0;两种方法t检验结果P值均大于0.05。采用一测多评法检测黄芩药材具有良好的精密度、稳定性、重复性与准确度,所得结果与外标法比较无显著差异,可用于黄芩药材质量控制。A quantitative analysis of multi-components by single marker method(QAMS)was established to simultaneous detect the content of baicalin,wogonoside,baicalein and wogonin in Scutellaria baicalensis.HPLC method was taken to detect the peak areas and retention times of baicalin,wogonoside,baicalein and wogonin,while baicalin was used as internal standard substance to calculate the relative correction factor and relative retention values.After that,the results were verified by different HPLC instruments.The results indicated that the relative correction factors of wogonoside,baicalein and wogonin were about 1.02,1.66,and 0.93,and the RSD values were 2.03%,3.25%,1.85%(n=15),respectively.The relative retention of wogonoside,baicalein and wogonin were about 2.11,2.80,and 3.57,while the RSD values were 0.83%、0.96%、0.98%(n=15),respectively.The RSD values of the precision,stability,repeatability,and different instrument validation tests were less than 3.5%(n=6).The error value of the experimental results over wogonoside,baicalein,and wogonin were about 0.53~0.70%,-1.30~3.33%,and-2.99~0 via QAMs and external standard method(ESM),respectively.And the P values of T-test results were greater than 0.05 for QAMs and ESM.In short,QAMs method had good precision,stability,repeatability and accuracy,which can be used for the quality control of scutellariae radix.
