摘  要:目的:观察通督启神针法对APP/PS1小鼠学习记忆行为及海马Aβ淀粉样沉积、炎症因子TNF-α的影响,探讨通督启神针法治疗AD的作用机制。方法:4月龄APP/PS1小鼠共16只,随机数字表达法分组,模型组8只,电针组8只。同龄野生型小鼠8只为对照组,以“百会”“印堂”“人中”3穴进行电针干预,每次30 min,1次/d,共4周。水迷宫实验检测各组小鼠干预前的学习记忆功能情况,干预后,新物体识别实验的偏好指数检测各组小鼠学习记忆功能的表现,Westernblot法测定海马区淀粉样蛋白(Aβ)、核因子激活的B细胞的κ-轻链增强(NF-κB)和肿瘤坏死因子-α蛋白(TNF-α)表达水平。结果:干预前,与对照组比较,在训练的第3、4天,模型组逃避潜伏期明显增加(P<0.01或P<0.05),穿越第3象限次数减少(P<0.01);各组小鼠的游泳速度差异无统计学意义。与对照组相比,实验第3、4天,干预前电针组逃避潜伏期明显增长(均P<0.01),穿越第3象限次数减少(P<0.01);而模型组与电针组之间差异无统计学意义。干预后与模型组比较,电针组对新物体偏好指数增加(P<0.01或P<0.05),脑内海马Aβ表达水平降低(P<0.01),海马中TNF-α和NF-κB分别显著降低(P<0.01或P<0.05)。结论:通督启神针法可通过抑制NF-κB通路、减少炎症因子TNF-α蛋白表达、减少Aβ沉积提高学习记忆,从而延缓AD的发展。Objective:To observe the effects of Tongdu Qishen acupuncture on learning and memory behavior,and hippocampal AβAmyloid deposition and flammatory factor of TNF-αin APP/PS1 mice,thus to explore the mechanism of Tongdu Qishen acupuncture in the treatment of AD.Methods:16 APP/PS1 mice of 4-month-old were randomly divided into the model group and the electro-acupuncture(EA)group,with 8 mice in each group.Another 8 wild-type mice of the same age were used as the blank control.EA intervention was applied to Baihui(DU20),Yintang(EX-HN3)and Renzhong(DU26),30 minutes each time,once a day for 4 weeks.The learning and memory function of mice was detected by water maze test before the intervention in each group.After the intervention,the preference index of new object recognition test was used to detect the performance of learning and memory function of mice in each group,and the expression levels of Aβ,NF-κB and TNF-αin the hippocampus were measured by Western blot.Results:Before the intervention,the escape latency increased significantly(P<0.05,P<0.01),the number of crossing the third quadrant decreased in the model group compared to that in the control group on the third and fourth days of training(P<0.01).There was no statistical difference in the swimming speed among the groups.Compared to that in the control group,the escape latency increased significantly(P<0.01),and the number of crossing the third quadrant decreased in the EA group on the third and fourth days before the intervention(P<0.01);of which there were no statistical differences between the model group and the EA group.After the intervention,the preference index for new objects increased(P<0.05),the expression of Aβin hippocampus decreased(P<0.01),and the hippocampal levels of TNF-αand NF-κB significantly decreased in the EA group compared to that in the model group(P<0.05,P<0.01).Conclusion:Tongdu Qishen acupuncture can reduce the inflammatory factor of TNF-αand decrease Aβdeposition by inhibiting NF-κB pathway to improve learning and memory,so
