摘 要:目的研究益智仁提取物延缓秀丽隐杆线虫衰老的功效及其作用机制。方法给予秀丽隐杆线虫一定浓度的益智仁提取液(20、40、80和100μg/mL),通过测定秀丽隐杆线虫N2野生型和DAF-16突变株型线虫的寿命,高浓度下N2后代数及线虫的抗高温逆性和抗氧化能力,评价了益智仁提取物延寿抗衰老的功效,并初步探讨了益智仁提取物延缓衰老的作用机制。结果实验结果表明,浓度100μg/L的益智仁提取物可以提高线虫寿命,增加线虫的高温抗逆性,增加CAT、SOD抗氧化生理指标的表达量以及DAF-16、SOD-3基因的表达量。结论益智仁提取物可以通过提高抗氧化酶活力和抗氧化基因的表达水平,延缓线虫衰老及急性热应激下的应激能力,延缓线虫衰老。Objective To investigate the effects and mechanism of Yizhiren(Alpinia oxyphylla)extract on delaying the aging of Caenorhabditis elegans.Methods The certain concentration of Yizhiren(Alpinia oxyphylla)extract(20μg/mL,40μg/mL,80μg/mL and 100μg/mL)was given to Caenorhabditis elegans.The effects of Yizhiren(Alpinia oxyphylla)extract on prolonging life and anti-aging were evaluated by measuring the life span of Caenorhabditis elegans N2 wild-type and daf-16 mutant nematodes,the post generation of N2 at high concentration,the resistance to high temperature stress and antioxidant capacity of nematodes,the anti-aging mechanism of Yizhiren(Alpinia oxyphylla)extract was discussed.Results The results showed that Yizhiren(Alpinia oxyphylla)extract(100μg/L)could prolong the life span of nematodes,improve the high temperature resistance of nematodes,increase the expressions of CAT and SOD antioxidant physiological indexes and the expressions of DAF-16 and SOD-3 genes.Conclusion The extract of Yizhiren(Alpinia oxyphylla)could delay the aging and stress ability of nematode under acute heat stress by improving the activity of antioxidant enzymes and the expression level of antioxidant genes.
