滋髓生血胶囊对再生障碍性贫血小鼠造血康复效果的影响

摘　　要：目的探讨滋髓生血胶囊对再生障碍性贫血小鼠造血康复效果的影响。方法研究选用雄性DBA小鼠60只,随机分为6组,每组10只:空白对照组无处理,再生障碍性贫血模型组接受再障模型后无处理;阳性药物对照组给予环孢素胶囊,司坦唑醇片,高剂量中药治疗组给予滋髓生血胶囊,120 mg/(kg·d),中剂量治疗组给予滋髓生血胶囊,60 mg/(kg·d),低剂量对照组给予滋髓生血胶囊,30 mg/(kg·d),治疗15 d为1个周期。检测小鼠的血细胞计数,采用流式法对外周血Th1/Th2/Th17细胞因子含量进行测定,统计骨髓有核细胞ERK蛋白的表达以及磷酸化的水平,统计骨髓造血祖细胞计数。结果与再生障碍贫血模型组小鼠相比,阳性药物对照组,3组中药治疗组的Hb浓度,WBC计数以及PLT计数均明显提升,与阳性药物对照组相比,低剂量组的差异不明显,中剂量、高剂量中药治疗组小鼠的提升更明显。与再生障碍贫血模型组小鼠相比,阳性药物对照组IFN-γ含量显著降低,低剂量中药治疗组显著高于模型组,高剂量中药治疗组显著低于模型组,3组中药治疗组均高于阳性药物治疗组,与再生障碍贫血模型组小鼠相比,低剂量、中剂量中药治疗组TNF明显提升,与阳性药物对照组小鼠相比,低剂量、中剂量中药治疗组TNF明显提升,与再生障碍贫血模型组小鼠相比,中剂量、高剂量中药治疗组IL-2含量明显提升,与阳性药物对照组小鼠相比,中剂量、高剂量中药治疗组IL-2明显提升,与再生障碍贫血模型组小鼠相比,高剂量中药治疗组IL-2含量显著降低,与再生障碍贫血模型组小鼠相比,中剂量、高剂量中药治疗组IL-4含量明显提升,与阳性药物对照组小鼠相比,中剂量、高剂量中药治疗组IL-4明显提升,与再生障碍贫血模型组小鼠相比,3组中药治疗组IL-6含量明显提升,与阳性药物对照组小鼠相比,3组中药治疗组IL-6明显提升,与再生障碍贫血�Objective To explore the effect of Zisui Shengxue Capsule(滋髓生血胶囊)on hematopoietic rehabilitation in aplastic anemia mice.Methods Sixty male DBA mice were selected in this study and randomly divided into six groups(n=10).The blank control group had no treatment,and the aplastic anemia model group had no treatment after receiving the aplastic anemia model.The positive drug control group was given Cyclosporine Capsule and Stanozolol Tablet.The high dose traditional Chinese medicine treatment group was given Zisui Shengxue Capsule[120 mg/(kg·d)].The medium dose treatment group was given Zisui Shengxue Capsule[60 mg/(kg·d)],and the low dose control group was given Zisui Shengxue Capsule[30 mg/(kg·d)],15 days as a cycle.The blood cell count of mice was detected,the contents of Th1/Th2/Th17 cytokines in peripheral blood was measured by flow cytometry,the expression of ERK protein and phosphorylation level of bone marrow nucleated cells were counted,and the number of bone marrow hematopoietic progenitor cells was counted.Results Compared with those in the aplastic anemia model group,the Hb concentration,WBC count and PLT count in the positive drug control group and three traditional Chinese medicine treatment groups were significantly increased.Compared with the positive drug control group,the difference in the low dose group was not significant.Obviously,the mice in the medium and high dose Chinese medicine treatment groups had more significant improvement.Compared with those in the aplastic anemia model group,the IFN-γcontent in the positive drug control group was significantly reduced.The content in the the low dose Chinese medicine treatment group was significantly higher than that in the the model group,and the content in the high dose Chinese medicine treatment group was significantly lower than that of the model group.The IFN-γcontents in three traditional Chinese medicine treatment groups were higher than that of the positive drug treatment group.Compared with that of the aplastic anemia model gr
