摘 要:目的研究五味子乙素对慢性铝中毒大鼠海马突触超微结构及学习与记忆功能的影响.方法用AlCl_(3)水溶液拌入饲料中连续喂养大鼠建立慢性铝中毒模型.按照随机分组方法,将慢性铝中毒动物模型的大鼠分为5组:模型组、阳性对照组和低、中、高3个剂量实验组.阳性对照组用吡拉西坦按40 mg·kg^(-1)灌胃给药;低、中、高3个剂量实验组用五味子乙素分别按10,20,40 mg·kg^(-1)进行灌胃;正常组不作任何处理.用Morris水迷宫定位航行实验测试大鼠学习与记忆能力,用透射电子显微镜观察大鼠海马突触超微结构的改变.结果正常组、模型组和高剂量实验组于第4天的潜伏期分别为(27.45±11.95),(48.85±9.42)和(32.35±6.20)s;这3组的海马CA1区突触数量分别为(44.80±3.42),(22.40±3.36)和(30.20±3.27)个;这3组的突触间隙宽度分别为(12.20±1.92),(21.40±4.83)和(13.40±2.07)nm;这3组的突触后致密物质厚度分别为(29.00±4.30),(17.00±2.92)和(23.60±3.44)nm.模型组的上述指标与正常组比较,差异均有统计学意义(均P<0.01);高剂量实验组的上述指标与模型组比较,差异均有统计学意义(均P<0.01).结论五味子乙素可以改善海马突触超微结构的变化以及改善突触结构参数的异常变化,从而提高学习记忆能力.Objective To study the effects of schisandrin B on the ultrastructure of hippocampal synapse as well as the learning and memory function in rats with chronic aluminum intoxication.Methods The model of chronic aluminum intoxication was established by feeding the rats continuously with the fodder miexed with AlCl_(3) aqueous solution.The rats of chronic aluminum poisoning animal model were divided into 5 groups by randomly methods:model group,positive control group and low-dose,middle-dose,high-dose experimental groups(experimental-L,experimental-M,experimental-H groups).The positive control group was given piracetam by intragastric administration at a dose of 40 mg·kg^(-1).The experimental-L,-M,-H groups were given intragastrically schisandrin B at doses of 10,20,and 40 mg·kg^(-1),respectively.The normal group was not treated.Thenavigation experiment in the Morris water maze was carried out to test the learning and memory ability of rats.The ultrastructural changes of hippocampal synapses were observed by transmission electron microscopy.Results The latency at forth day of normal group,model group and experimental-H group were(27.45±11.95),(48.85±9.42)and(32.35±6.20)s,respectively;the number of synapses in the CA1 region of the hippocampus in the three groups were(44.80±3.42),(22.40±3.36)and(30.20±3.27)count,respectively;their widths of synaptic gap in the three groups were(12.20±1.92),(21.40±4.83)and(13.40±2.07)nm,respectively;the thickness of post-synaptic dense material in the three groups were(29.00±4.30),(17.00±2.92)and(23.60±3.44)nm,respectively.Compared between model group and normal group,the difference of the above indexes were significant(all P<0.01).Compared between experimental-H and model group,the difference of the above indexes were significant(all P<0.01).Conclusion Schisandrin B can improve the changes in the ultrastructure of hippocampal synapses as well as the abnormal changes in parameters of synaptic structure,thereby enhancing the ability of learning and memory.
