摘  要:目的研究摄食抑制因子(Nesfatin-1)对2型糖尿病(T2DM)模型大鼠中胰岛素的影响。方法用高糖高脂饲料喂养SD大鼠和腹腔注射链脲佐菌素制备T2DM模型大鼠。将模型大鼠随机分为模型组和低、高剂量实验组,每组3只;另取9只正常大鼠随机分为空白组、低剂量对照组和高剂量对照组,每组3只。空白组和模型组在第三脑室注射人工脑脊液(aCSF)10μL;低剂量对照组和低剂量实验组注射Nesfatin-1(5 pmol)10μL,高剂量对照组和高剂量实验组注射Nesfatin-1(50 pmol)10μL。用酶联免疫吸附法检测胰岛素含量,用实时荧光定量聚合酶链反应检测蛋白激酶B(AKT)、磷酸烯醇式丙酮酸激酶(PEPCK)和葡萄糖-6-磷酸酶(G-6-pase)mRNA的表达水平。结果低、高剂量对照组和低、高剂量实验组及模型组的血清胰岛素含量分别为(24.39±1.44),(22.95±1.90),(19.51±0.97),(20.28±1.43)和(17.84±2.33)mU·L^(-1),AKT mRNA表达量分别为0.90±0.11,0.91±0.27,0.71±0.16,0.83±0.01和0.48±0.07,PEPCR mRNA表达量分别为0.87±0.14,0.76±0.08,0.60±0.17,0.86±0.14和0.44±0.13,G-6-pase mRNA表达量分别为1.07±0.13,1.01±0.12,0.69±0.13,1.06±0.12和0.39±0.09。模型组的上述指标与高剂量实验组比较,差异均有统计学意义(均P<0.05)。结论Nesfatin-1通过促进AKT、PEPCK和G-6-Pase通路的表达,进而改善胰岛素抵抗。Objective To study the effect of Nesfatin-1 on insulin in type 2 diabetes mellitus(T2DM)rats.Methods T2DM model rats were established by feeding with high-sugar and high-fat diet and intraperitoneal injection of streptozotocin(STZ).The model rats were divided into model group and experimental-L,-H groups with 3 cases per group.Another 9 normal rats were divided into blank group and control-L,-H groups with 3 cases per group.The blank and model groups were injected with artificial cerebrospinal solution(a CSF)10μL in the third ventricle.The control-L and experimental-L groups were injected with Nesfatin-1(5 pmol)10μL.The control-H and experimental-H groups were injected with Nesfatin-1(50 pmol)10μL.Insulin was determined by enzyme-linked immunosorbent assay.The expression of protein kinase B(AKT),phosphoenolpyruvate kinase(PEPCK)and glucose-6-phosphatase(G-6-pase)mRNA was detected by reverse transcription-polymerase chain reaction.Results The serum insulin contents in control-L,-H groups,experimental-L,-H groups and model groups were(24.39±1.44),(22.95±1.90),(19.51±0.97),(20.28±1.43)and(17.84±2.33)m U·L^(-1),the expression levels of AKT mRNA were0.90±0.11,0.91±0.27,0.71±0.16,0.83±0.01 and 0.48±0.07,the expression levels of PEPCR mRNA were0.87±0.14,0.76±0.08,0.60±0.17,0.86±0.14 and 0.44±0.13,the expression levels of G-6-pase mRNA were 1.07±0.13,1.01±0.12,0.69±0.13,1.06±0.12 and 0.39±0.09.There were statistically significant differences between model group and experimental-H group.Conclusion Nesfatin-1 improves the insulin resistance by promoting the expression of AKT,PEPCK and G-6-pase pathways.
