摘 要:通过线粒体D-loop基因部分序列对广东惠东海龟国家级自然保护区人工繁育的4个群体共99只绿海龟样本进行扩增测序及分析。研究结果表明,绿海龟人工繁育4个群体的D-loop基因片段序列的A+T含量为68.1%,碱基组成具有明显的偏向性;共定义了3个单倍型,单倍型多样性指数在0.000~0.212之间,核苷酸多样性指数在0.00000~0.00111之间;人工繁育4个群体的遗传分化系数(F_(st))值均大于0.05,4个群体中有3个群体之间不存在基因交流现象;采用邻位连接法(NJ)对99个序列片段构建了系统发育树,G组群体单独聚为一分支,C组、H组和LM组群体聚为另一分支。研究结果说明,广东惠东海龟国家级自然保护区绿海龟人工繁育的各个群体内遗传多样性差异较小,群体间存在较大遗传分化,绿海龟人工繁育4个群体仍具有较高的遗传多样性。研究结果为建立相应的绿海龟人工繁育遗传谱系以及开展绿海龟种质资源保护奠定理论基础。The partial sequences of mitochondrial D-loop gene were used to amplify the sequencing and analyze a total of 99 green sea turtle(Chelonia mydas)samples from four cultured populations in National Huidong Sea Turtle Nature Reserve.The results showed that A+T content of D-loop gene fragments of the four cultured populations was 68.1%and the base composition showed obvious bias.A total of three haplotypes were defined,with haplotype diversity ranging from 0.000 to 0.212 and nucleotide diversity ranging from 0.00000 to 0.00111.The genetic differentiation coefficient(F_(st))of the four cultured populations were all greater than 0.05 and there was no gene flow(N_(m))between three of the four cultured populations.According to the neighbor-joining tree of 99 D-loop partial sequences,the first clade only consist of the G group population and the second clade consist of the C,H and LM group populations.The result indicated that there was low level of genetic differentiation in every cultured population and high level of genetic differentiation between the four cultured populations in National Huidong Sea Turtle Nature Reserve.This study lays the theoretical foundation for the establishment of the corresponding genetic genealogy of green sea turtles and the conservation of green sea turtle germplasm resources.
