摘  要:蚯蚓是土壤中生物量最大的无脊椎动物,是土壤环境污染研究良好的实验材料,蚯蚓体腔细胞可以进行多种细胞毒理实验。实验将蚯蚓体腔细胞用于过氧化氢(H_(2)O_(2))构建的细胞损伤模型研究,为将这种实验材料用于初步筛选对细胞有保护作用的天然抗氧化剂的探索提供参考。用单细胞凝胶电泳技术分析不同浓度H_(2)O_(2)对蚯蚓离体体腔细胞DNA的损伤作用。随H_(2)O_(2)浓度增大,代表DNA损伤程度的尾部DNA含量(T_(DNA)%)、尾长(T_(L))、尾矩(T_(M))和Olive尾矩(T_(OM))4个参数值也随之升高,表明H_(2)O_(2)对细胞DNA造成了损伤,且浓度越高,细胞DNA损伤越严重,两者呈显著的剂量—效应关系。实验揭示了蚯蚓体腔细胞对H_(2)O_(2)敏感,可用于初步筛选天然抗氧化剂的研究。As the invertebrate with the largest biomass in soil,earthworms are good experimental materials for soil environmental pollution research.Moreover,earthworm coelomocytes can be used for a variety of cytotoxicological experiments.In this experiment,earthworm coelomocytes—those which provide a reference for the preliminary screening of natural antioxidants with protective effects on cells—were used to construct a cell injury model induced by hydrogen peroxide.Single-cell gel electrophoresis was used to analyze damage effect of different concentrations of H_(2)O_(2) on DNA of isolated earthworm coelomocytes in vitro.The values of T_(DNA)%,T_(L),T_(M) and T_(OM),which represent the degree of DNA damage,increased with the increase of H_(2)O_(2) concentration,indicating that H_(2)O_(2) caused damage to earthworm coelomocytes.There was a significant dose-effect relationship between H_(2)O_(2) concentration and DNA damage of coelomocytes,i.e.the higher the H_(2)O_(2) concentration,the more serious DNA damage of coelomocytes.The experimental results showed that earthworm coelomocytes are sensitive to H_(2)O_(2) and can be further used for preliminary screening of natural antioxidants.
