摘 要:目的:利用网络药理学方法探讨血府逐瘀汤治疗继发性闭经的潜在作用机制。方法:利用中药系统药理学数据库和分析平台(TCMSP)以及ETCM数据库查找出血府逐瘀汤中所有药物的活性成分,通过DrugBank数据库、DisGeNET数据库、GeneCards数据库查找继发性闭经相关靶点,利用Cytoscape 3.8.0软件构建血府逐瘀汤中所有中药的活性成分及对应靶点(活性成分-靶点)网络。利用STRING网络数据库构建及扩增血府逐瘀汤与继发性闭经之间的交集靶点蛋白相互作用(PPI)网络,使用Cytoscape 3.8.0软件绘制PPI网络图。通过DAVID数据库对核心靶点进行基因本体(GO)分类富集分析和京都基因与基因组百科全书(KEGG)通路富集分析。结果:筛选出163个血府逐瘀汤作用靶点,其中与继发性闭经相关的作用靶点有34个,扩增以后一共得到64个相关靶点。血府逐瘀汤主要通过作用于白细胞介素-6、肿瘤抑制因子P53等靶点,调控类固醇功能、肿瘤信号通路等多个生物过程及通路,来发挥治疗继发性闭经的作用。结论:血府逐瘀汤可作用于继发性闭经患者的多个靶点,参与多条信号通路的调控。Objective: To explore the potential mechanism of Xuefu Zhuyu tang for secondary amenorrhea based on network pharmacology. Methods:The Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP) and ETCM database were used to search active ingredients of all medicines in Xuefu Zhuyu tang;DrugBank database, DisGeNET database and GeneCards database were used to search related targets of secondary amenorrhea;Cytoscape 3.8.0 software was used to construct the network of active ingredients of all Chinese herbs in Xuefu Zhuyu tang and corresponding targets(active ingredients-targets). The target protein-protein interaction(PPI) network between Xuefu Zhuyu tang and secondary amenorrhea was constructed and expanded by the database of STRING website, and the PPI network diagram was drawn by Cytoscape 3.8.0 software. Gene ontology(GO) classification enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG) pathway enrichment analysis were performed on the core targets through DAVID database. Results:A total of 163 targets of Xuefu Zhuyu tang were screened out,among which 34 targets were related to secondary amenorrhea,and a total of 64 related targets were obtained after amplification. Mainly by acting on interleukin-6,tumor suppressor P53 and other targets to regulate the function of steroids,tumor pathways and other biological processes and pathways,Xuefu Zhuyu tang plays a role in the treatment of secondary amenorrhea. Conclusion:Xuefu Zhuyu tang can act on multiple targets of secondary amenorrhea and participate in the regulation of multiple signaling pathways.
