摘 要:目的 研究针刺“曲池”“足三里”联合穴位贴敷“脾俞”对高血压前期盐敏感大鼠肾脏水通道蛋白1(AQP1)、水通道蛋白2(AQP2)表达的影响,从而探讨针刺“曲池”“足三里”联合穴位贴敷“脾俞”的降压机制。方法 盐抵抗大鼠(DR)8只作为空白组;盐敏感大鼠(DS)大鼠32只,随机分为模型组、针刺组、穴位贴敷组和针刺+穴位贴敷组,每组8只。大鼠统一在SPF动物实验中心适应性喂养1周后,改为8%高盐饲料喂养造高血压前期模型。针刺组取双侧“足三里”“曲池”穴施针,1次/天,20 min/次,6次/周,治疗4周。穴位贴敷组取“脾俞”给予中药贴敷,6 h/天,治疗时间与针刺组相同。针刺+穴位贴敷组在针刺后进行穴位贴敷,治疗方法与时间同针刺组与穴位贴敷组。测量治疗期间大鼠血压变化;ELISA法检测血清肾素血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)含量;RT-PCR检测大鼠肾组织水通道蛋白1(AQP1)、水通道蛋白2(AQP2)mRNA表达;免疫组化法检测大鼠肾组织水通道蛋白1、水通道蛋白2表达。结果 治疗后,大鼠血压下降。与空白组比较,模型组AngⅡ、ALD、AQP1、AQP2含量明显增加(P<0.05);与模型组比较,针刺组、穴位贴敷组、针刺+穴位贴敷组AngⅡ、AQP1含量明显降低(P<0.05),针刺组和针刺+穴位贴敷组ALD、AQP2明显降低(P<0.05)。结论 针刺联合穴位贴敷能有效降低高血压前期盐敏感大鼠血压,降低AngⅡ、ALD、AQP1、AQP2表达,改善机体水液代谢紊乱,这可能是针刺联合穴位贴敷调节盐敏感性高血压前期大鼠血压的机制之一。Objective To study the effects of acupuncture "Quchi(LI11)" and "Zusanli(ST36)" combined with acupoint application "Pishu(BL20)" on AQP1 and AQP2 in the kidneys of salt-sensitive rats with prehypertension, so as to explore the effects of acupuncture at "Quchi(LI11)" and "Zusanli(ST36)" the blood pressure reduction mechanism of "Pishu(BL20)" combined with acupoint application. Methods Eight salt-resistant rats(DR) were regarded as the normal group;32 salt-sensitive rats(DS) rats were randomly divided into model group, acupuncture group, acupoint application group and acupuncture + acupoint application group. 8 per group. Rats were uniformly adaptively fed for one week in the SPF animal experimental center, then used high-salt feed to create a prehypertensive model. In the acupuncture group, acupuncture was given at the acupoints "Zusanli(ST36)" and "Quchi(LI11)", once a day, 6 times a week, for 4 weeks. In the acupoint application group, "Pishu(BL20)" was applied with traditional Chinese medicine, 6 h/d, and the treatment time was the same as that of the acupuncture group. In the acupuncture and acupoint application group, acupoint application was performed after acupuncture, and the treatment method and time were the same as the acupuncture group and the acupoint application group. The changes in blood pressure of rats during treatment was measured;ELISA method was used to detect serum renin angiotensin Ⅱ(Ang Ⅱ), aldosterone(ALD) content;RT-PCR was used to detect rat kidney tissue aquaporin 1(AQP1), aquaporin 2(AQP2) MRNA expression;Immunohistochemical method was used to detect the expression of aquaporin 1(AQP1) and aquaporin 2(AQP2) in rat kidney tissue.Results After treatment, the blood pressure of the rats decreased. Compared with the normal group, the content of AngⅡ, ALD,AQP1, and AQP2 in the model group was significantly increased(P< 0.05);Compared with the model group, the acupuncture group, acupoint application group, acupuncture + acupoint application group AngⅡ, AQP1 the content was significant
