摘 要:目的检测微小RNA-760(miR-760)、细胞因子信号传导抑制因子3(SOCS3)在上皮性卵巢癌患者血清中的表达,分析其临床意义。方法选取2015年3月至2017年5月苏州市吴中人民医院及苏州大学附属第二医院诊治的68例上皮性卵巢癌患者作为研究对象,设为试验组。另随机选取同期诊治的68例卵巢良性肿瘤患者作为对照组。采用实时荧光定量聚合酶链反应法检测两组血清miR-760表达;采用酶联免疫吸附试验法检测两组血清SOCS3表达;采用Pearson法分析上皮性卵巢癌患者血清miR-760与SOCS3表达的相关性;对治疗后的上皮性卵巢癌患者进行随访,记录患者生存状态;采用Kaplan-Meier法生存曲线分析miR-760、SOCS3表达与上皮性卵巢癌预后的关系;采用COX回归分析影响上皮性卵巢癌预后的危险因素。结果与对照组比较,试验组血清miR-760表达显著升高,SOCS3表达显著降低,差异具有统计学意义(P<0.05);上皮性卵巢癌患者血清miR-760、SOCS3表达与临床分期、淋巴结转移、组织分级有关(P<0.05);上皮性卵巢癌患者血清miR-760与SOCS3表达呈负相关(r=-0.706,P<0.05);随访期间,miR-760高表达组生存率(27.0%)低于低表达组(58.1%),SOCS3高表达组生存率(56.3%)高于低表达组(27.8%),差异具有统计学意义(P<0.05);多因素COX分析表明,临床分期、淋巴结转移、组织分级、miR-760表达是影响上皮性卵巢癌预后的危险因素,SOCS3是保护因素(P<0.05)。结论miR-760、SOCS3表达与上皮性卵巢癌发生发展及预后均有关,可能是上皮性卵巢癌发病机制的潜在研究靶点。Objective To detect the expressions of miR-760 and suppressor of cytokine signaling 3(SOCS3)in serum of patients with epithelial ovarian cancer and analyze their clinical significance.Methods A total of 68 cases of epithelial ovarian cancer patients diagnosed in Wuzhong People′s Hospital of Suzhou and Second Affiliated Hospital of Suzhou University from March 2015 to May 2017 were selected as the research objects,and 68 cases of benign ovarian tumor were selected as the control group.Real-time quantitative PCR was used to detect the expression of miR-760 in serum of the two groups.The expression of serum SOCS3 was detected by enzyme-linked immunosorbent assay.Pearson method was used to analyze the correlation between serum miR-760 and SOCS3 levels in patients with epithelial ovarian cancer.Patients with epithelial ovarian cancer were followed up after treatment and their survival status was recorded.The relationship between miR-760,SOCS3 and prognosis of epithelial ovarian cancer was analyzed by Kaplan-Meier survival curve.COX regression analysis was used to analyze the risk factors of epithelial ovarian cancer.Results Compared with those in the control group,the expression of miR-760 was significantly higher,and the expression of SOCS3 was significantly lower(P<0.05).The expression of serum miR-760 and SOCS3 were correlated with clinical stage,lymph node metastasis and histological grade(P<0.05).There was a negative correlation between serum miR-760 and SOCS3 expression levels in patients with epithelial ovarian cancer(r=-0.706,P<0.05).During the follow up,the survival rate of miR-760 high expression group(27.0%)was lower than that of low expression group(58.1%),and the survival rate of SOCS3 high expression group(56.3%)was higher than that of low expression group(27.8%).Multivariate COX analysis showed that clinical stage,lymph node metastasis,histological grade and miR-760 expression were risk factors for the prognosis of epithelial ovarian cancer,and SOCS3 was a protective factor(P<0.05).Conclusions MiR-760
