苗药五藤膏外敷对膝骨性关节炎兔软骨RANK、RANKL及Notch4受体表达的影响

摘　　要：目的:探讨苗药五藤膏外敷对膝骨性关节炎(KOA)兔软骨RANK、RANKL及Notch4受体表达的影响。方法:将96只健康新西兰大白兔随机分为空白对照组,模型对照组,苗药五藤膏高、中、低剂量组以及阳性药物组,每组16只,除空白对照组外,其余各组建立兔KOA软骨模型,分别于用药1、2、3、4周取患侧兔关节软骨,切片后进行HE染色,采用RT-PCR法检测兔RANK、RANKL、Notch4受体表达量,并将第4周切片结果进行病理学Mankin评分。结果:与空白对照组比较,模型对照组Mankin评分显著升高(P<0.05),各治疗组Mankin评分均显著低于模型对照组(P<0.05);与模型对照组比较,各治疗组RANK、RANKL、Notch4受体表达有不同程度的降低(P<0.05)。结论:苗药五藤膏能够降低KOA模型兔软骨RANK、RANKL、Notch4受体表达,其中五藤膏高剂量组降低效果更明显。Objective: To investigate the effects of Miao medicine Wuteng Ointment on the expression of RANK, RANKL and Notch4 receptors in KOA rabbit cartilage by external application of Miao medicine Wuteng Ointment. Methods: A total of 96 healthy New Zealand white rabbits were randomly divided into blank control group, model control group, Miao medicine Wuteng Ointment high, medium and low dose treatment groups, and flurbiprofen Papua cream treatment group, each group of 16, in addition to the blank control group, the rest of the group to establish a rabbit model of KOA cartilage, medicine 1, 2, 3, 4 weeks respectively the articular anatomy, and executed a batch of animals of rabbit articular cartilage, slice after HE staining, detection by RT-PCR method in rabbit RANK, RANKL, Notch4 receptor expression, and pathology Mankin slice 4 weeks for results. Results: Compared with the blank control group, the model control group’s Mankin scores significantly increased(P<0.05). The scores of treatment groups significantly decreased than that of the model control group(P<0.05). Compared with the model control group, the expressions of RANK, RANKL and Notch4 receptors in rabbits in treatment groups were decreased to different degrees(P<0.05). Conclusion:Miao medicine Wuteng Ointment can reduce the expression of RANK, RANKL and Notch4 receptors in KOA model rabbit cartilage,and the reduction effect is more obvious in the high-dose group of Miao medicine Wuteng Ointment.
