摘 要:目的:探讨雄蚕益肾方对肝郁肾虚阳痿大鼠性腺轴及睾丸P450scc、StAR、NR5A1表达的影响。方法:50只8周龄SPF级雄性SD大鼠随机分为空白组、模型组、他达拉非组、疏肝益肾组和雄蚕益肾方低、中、高剂量组,采用氢化可的松注射液肌注加束缚四肢复合造模方法连续14d诱发肝郁肾虚阳痿模型。造模成功后,他达拉非组给予他达拉非片溶液0.52mg·kg^(-1)·d^(-1)灌胃,疏肝益肾组给予疏肝益阳胶囊溶液0.3125g·kg^(-1)·d^(-1)灌胃,雄蚕益肾方低、中、高剂量组给予10.4、20.8、41.6g·kg^(-1)·d^(-1)雄蚕益肾方溶液灌胃,空白组、模型组给予等体积蒸馏水灌胃,各组每日均早、晚各灌胃1次,连续28d。ELISA法测定血清中睾酮(T)、促黄体生成素(LH)、卵泡刺激素(FSH)含量,HE染色光镜下观察睾丸微细结构,免疫组化法测定睾丸P450scc、StAR、NR5A1表达情况。结果:与空白组比较,模型组大鼠血清中T含量及睾丸中P450scc、StAR、NR5A1表达均显著降低(P<0.05)。与模型组比较,疏肝益阳组和雄蚕益肾方各剂量组血清T及睾丸中P450scc表达均显著升高(P<0.05);雄蚕益肾方各剂量组大鼠睾丸中StAR表达均显著升高(P<0.05),血清FSH水平均显著降低(P<0.05);雄蚕益肾方高、中剂量组大鼠睾丸中NR5A1表达显著升高(P<0.05)。结论:雄蚕益肾方可调节性腺轴和提高P450scc、StAR、NR5A1表达。Objective: To study the effects of the Xiongcan Yishen Formula on the expression of the gonadal axis and the testis P450scc, StAR, NR5A1 in the rats with liver depression, kidney deficiency and impotence. Methods: A total of 50 8-week-old SPF class male SD rats were randomly divided into blank control group, model group, western drug control group, Chinese patent medicine control group and Xiongcan Yishen Formula low, medium and high dose groups, 10 rats were randomly selected randomly from 50 male SD rats as the normal control rats, and the rest rats were induced by hydrocortisone injection combined with binding limbs for 14 days to induce hepatic deficiency of kidney deficiency. After successful modeling, 5 in each group, the control group of western medicine was given Tadarafifen tablet solution 0.52 mg·kg^(-1)·d^(-1) for gavage, and the Chinese patent medicine control group was given 0.3125 g·kg^(-1)·d^(-1), the low, middle and high doses of Xiongcan Yishen Formula was 10.4, 20.8,41.6 g·kg^(-1)·d^(-1), the blank control group and the model group were given the same volume distilled water, each group was given intragastrically early and late every day for 28 days. The contents of T, LH and FSH in serum were determined by ELISA, the testicular HE staining was used to observe testicular microstructure under light microscope. Expressions of P450scc, StAR and NR5A1 were detected by immunohistochemical method. Results: Compared with blank group, the content of T in the serum and the expressions of P450scc, StAR and NR5A1 in testicular tissue of the model group decreased significantly(P<0.05). Compared with the model group, the serum T and the expression of P450scc in testicular tissue of Shugan Yiyang group and the Xiongcan Yishen Formula three dose groups increased significantly(P<0.05), the expression of StAR in testicular tissue of the Xiongcan Yishen Formula three dose groups was significantly increased(P<0.05) and the level of serum FSH was significantly decreased(P<0.05), the expression of NR5A1 in the
