摘 要:目的:基于胰岛素通路应用磷酸化抗体蛋白芯片技术探索益肾喘宁汤治疗肾气虚哮喘的作用机制。方法:将24只清洁级Wistar大鼠随机分为正常组、模型组、中药组,每组8只,分别予以相应措施进行处理,之后取出各组大鼠肺组织,运用磷酸化抗体蛋白芯片技术检测各组大鼠胰岛素通路相关位点蛋白的磷酸化水平,筛选出组间比较具有显著差异的蛋白。结果:与正常组比较,模型组大鼠IRS1、ELK1、GSK3β、EIF4EBP1、RPS6KB1、PRKAA1、BAD、LIPE等磷酸化水平均有所变化;用药后,IRS1、GSK3β、RPS6KB1、PRKAA1等的指标明显升高。结论:通过磷酸化抗体芯片分析,筛选出了益肾喘宁汤干预后哮喘大鼠胰岛素通路中磷酸化水平具有显著差异的位点蛋白,为进一步探讨其作用机制指明了方向。Objective:Based on insulin pathway, using phosphorylated antibody protein chip technology to explore the mechanism of Yishen Chuanning Decoction(YSCND) in the treatment of kidney qi deficiency asthma. Methods: A total of 24 clean Wistar rats were randomly divided into normal group, model group and YSCND group(n=8), and conducted with corresponding measures respectively. Then the lung tissues of rats in each group were taken out, and the phosphorylation level of proteins related to insulin pathway was detected by phosphorylated antibody protein chip technique, and the proteins with significant differences among groups were screened. Results: Compared with the normal group, the phosphorylation levels of IRS1, ELK1, GSK3β, EIF4 EBP1, RPS6 KB1, PRKAA1, BAD and LIPE in the model group changed, and the indexes such as IRS1, GSK3β, RPS6 KB1 and PRKAA1 increased significantly after treatment. Conclusion: Through the analysis of phosphorylated antibody chip, the site proteins with significant difference in phosphorylation level of insulin pathway in asthmatic rats treated with YSCND were screened, which pointed out the direction for further exploring its mechanism.
