摘 要:目的:探究戊戌年运气方静顺汤对慢性不可预见性温和刺激(CUMS)联合孤养所致抑郁模型大鼠行为学及不同脑区单胺类神经递质水平的影响。方法:30只SD雄性大鼠随机分为空白组、模型组、氟西汀组、柴芩温胆汤组、静顺汤组。采用CUMS联合孤养方法建立抑郁模型大鼠,造模同时给予药物刺激,42d后通过强迫游泳及悬尾实验进行行为学评价,采用高效液相色谱-电化学检测方法(HPLC-ECD)测定大鼠前额叶皮质、下丘脑单胺类神经递质的变化。结果:与模型组比较,柴芩温胆汤组、静顺汤组的强迫游泳和悬尾实验不动时间均显著缩短(P<0.01),静顺汤组前额叶皮质多巴胺(DA)显著升高(P<0.01),5-羟色胺(5-HT)显著升高(P<0.05),下丘脑3,4-二羟基苯乙酸(DOPAC)、高香草酸(HVA)显著升高(P<0.01)。结论:静顺汤能改善抑郁模型大鼠行为学症状,静顺汤发挥抗抑郁作用可能与提高前额叶皮质5-HT和DA水平、下丘脑的DOPAC和HVA水平有关。Objective: To compare the effects of Wuxu Year Yunqi Formula Jingshun Decoction and conventional antidepressant Chaiqin Wendan Decoction on behavior of depression model rats induced by chronic unpredictable mild stimulation(CUMS) combined with solitary rearing, and explore its mechanism of action. Methods: Thirty male SD rats were randomly divided into blank group, model group, fluoxetine group, Chaiqin Wendan Decoction group and Jingshun Decoction group. The depression model rat was established by CUMS combined with orphanage, and the model was given drug stimulation at the same time. After 42 days, the behavior was evaluated by forced swimming and tail suspension test. The changes of monoamine neurotransmitters in prefrontal cortex and hypothalamus were determined by HPLC-ECD. Results: Compared with the model group, the immobility time of forced swimming and tail suspension test in Chaiqin Wendan Decoction group and Jingshun Decoction group were significantly shorter(P<0.01), and the immobility time in Jingshun Decoction group was less than that in Chaiqin Wendan Decoction group, DA in the prefrontal cortex of the Jingshun Decoction group was significantly increased(P<0.01), 5-HT was significantly increased(P<0.05), and DOPAC and HVA in the hypothalamus were extremely increased(P<0.01). Conclusion: Jingshun Decoction can improve the behavioral symptoms of depression model rats, and its effect is more significant than that of Chaiqin Wendan Decoction. The antidepressant effect of Jingshun Decoction may be related to the increase of 5-HT and DA in the prefrontal cortex and the DOPAC and HVA levels in the hypothalamus.
