摘 要:目的:探究青蒿-鳖甲治疗系统性红斑狼疮(SLE)小鼠的效果,并从肠道菌群角度分析可能的机制。方法:运用Illumina MiSeq高通量测序技术检测MRL/lpr小鼠的肠道菌群结构,利用LEfSe分析挖掘青蒿-鳖甲改变的肠道菌群种类,采用Spearman方法对狼疮指标及改变的肠道菌群进行关联性分析。结果:高剂量青蒿-鳖甲治疗MRL/lpr小鼠的效果与激素相当,其能通过增加肠道菌群Alpha多样性指数,上调与狼疮病情显著负相关的菌属(Bacteroides、Parabacteroides和Odoribacter)和下调与狼疮病情显著正相关的菌属(Clostridium和Ruminococcus),达到治疗狼疮的目的。结论:青蒿-鳖甲治疗SLE时显著改变的肠道菌群,利于狼疮病情的缓解。Objective: To explore the therapeutic effect of Qinghao-Biejia in the treatment of MRL/lpr mice and reveal the possible mechanism from the perspective of gut microbiome. Methods: Illumina MiSeq sequencing was used to explore the microbial communities of MRL/lpr mice. LEfSe analysis was used to reveal the significantly altered gut microbiota by Qinghao-Biejia and Spearman method was employed to correlate the altered gut microbiota and lupus indices. Results: The therapeutic effect of high-dose Qinghao-Biejia was consistent with that of glucocorticoid. In addition, Qinghao-Biejia could increase Alpha diversity and negatively lupus activity-related microbial genus including Bacteroides, Parabacteroides and Odoribacter, and decrease positively lupus activity-related microbial genus including Clostridium and Ruminococcus. Conclusion: The significantly altered gut microbiota genus by Qinghao-Biejia is beneficial to the remission of lupus.
