摘 要:目的:研究枳实芍药散对便秘型肠易激综合征(C-IBS)模型大鼠结肠组织水通道蛋白3(AQP3)表达的影响,探讨枳实芍药散对C-IBS的作用及机制。方法:SPF级Wistar成年雄性大鼠60只,随机分为空白组10只和实验组50只。实验组大鼠采用0~4℃0.9%氯化钠溶液灌胃的方法复制C-IBS模型。模型制备成功后,实验组大鼠随机分为模型对照组,匹维溴铵组,枳实芍药散高、中、低剂量组,每组10只。空白组和模型对照组给予常温0.9%氯化钠溶液(10mL/kg)灌胃;匹维溴铵组给予匹维溴铵混悬液(13.5mg/kg)灌胃;枳实芍药散高、中、低剂量组给予枳实芍药散(3.28、1.64、0.82g/kg)灌胃,各14d。计算并比较各组大鼠的粪便含水量及腹壁撤退反射(AWR)评分。采用荧光定量PCR法检测AQP3 mRNA的表达,采用免疫组织化学染色法检测各组大鼠AQP3蛋白表达。结果:模型对照组大鼠粪便含水量较空白组显著降低(P<0.01),匹维溴铵组,枳实芍药散高、中剂量组较模型对照组粪便含水量显著升高(P<0.01)。在扩张压力为40、60、80mmHg时,模型对照组大鼠AWR评分较空白组显著升高(P<0.01);匹维溴铵和枳实芍药散高剂量组AWR评分较模型对照组显著下降(P<0.05,P<0.01)。模型对照组大鼠结肠组织AQP3 mRNA和蛋白的表达水平较空白组显著下降(P<0.05,P<0.01);匹维溴铵组、枳实芍药散高剂量组AQP3 mRNA和蛋白的表达水平较模型对照组显著升高(P<0.01,P<0.05)。结论:枳实芍药散可以治疗C-IBS,以高剂量疗效最佳,其机制可能与调节结肠组织中AQP3的表达有关。Objective: To study the effect of Zhishi Shaoyao Powder on the expression of aquaporin 3(AQP3) in colon tissue of constipation-predominant irritable bowel syndrome(C-IBS) model rats, and to explore the effect and mechanism of Zhishi Shaoyao Powder on C-IBS. Methods: Sixty adult male SPF Wistar rats were randomly divided into normal group(n=10) and experimental group(n=50). Rats in the experimental group were fed with 0.9% sodium chloride solution at 0~4℃ to establish the C-IBS model. After the model was successfully prepared, the rats in the experimental group were randomly divided into model control group, pinaverium bromide group, high, medium and low dose of Zhishi Shaoyao Powder groups, with 10 rats in each group. The normal group and the model control group were given 0.9% sodium chloride solution(10 mL/kg) at room temperature by gavage, the pinaverium bromide group was given pinaverium bromide(13.5 mg/kg) by gavage, the high, medium and low dose of Zhishi Shaoyao Powder groups were given Zhishi Shaoyao Powder(3.28, 1.64, 0.82 g/kg) by gavage. They were continuously administered for 14 d. The fecal water content and the score of abdominal withdrawal reflex(AWR) were calculated and compared. The expression of AQP3 mRNA was detected by qRT-PCR, while the expression of AQP3 protein was detected by immunohistochemical staining. Results: The fecal water content of rats in the model control group was significantly decreased than that in the normal group(P<0.01), and the fecal water content in the groups of pinaverium bromide, high and medium dose of Zhishi Shaoyao Powder was significantly increased than that in the model control group(P<0.01). Under 40, 60, 80 mmHg pressure, the AWR score of rats in the model control group was significantly increased than that in the normal group(P<0.01),and the AWR score in the groups of pinaverium bromide and high dose of Zhishi Shaoyao Powder was significantly decreased than that in the model control group(P<0.05, P<0.01).
