摘 要:目的:观察泽兰对肾小球内皮细胞(glomerular endothelial cells,GECs)糖萼核心蛋白的影响,揭示泽兰缓解高糖诱导GECs损伤的机制。方法:高糖环境下培养大鼠GECs,予泽兰及厄贝沙坦含药血清处理2 d,收集处理后的细胞,Western blot检测大鼠GECs糖萼核心蛋白Syndecan-1、Glypican-1和CD31、α-SMA表达,RT-PCR法检测mRNA水平。结果:高糖环境培养下,大鼠GECs糖萼核心蛋白Syndecan-1、Glypican-1、CD31及其mRNA表达下降,α-SMA及其mRNA表达增加(P<0.05);而泽兰及厄贝沙坦含药血清则可上调Syndecan-1、Glypican-1、CD31及其mRNA表达(P<0.05),抑制α-SMA及其mRNA表达(P<0.05);泽兰对Syndecan-1、Glypican-1的保护作用优于厄贝沙坦且呈剂量依赖性(P<0.05),厄贝沙坦对α-SMA的抑制作用更显著(P<0.05)。结论:泽兰通过保护糖萼核心蛋白缓解高糖诱导的GECs损伤,为泽兰治疗DN提供了又一重要理论依据。Objective:To observe the effects of Lycopus lucidus Turcz on glycocalyx core proteins of glomerular endothelial cells(GECs)and to reveal the mechanism of Lycopus lucidus Turcz alleviating GECs damage induced by high glucose.Methods:Rat GECs,cultured in high glucose environment were treated with drug-containing serum of Lycopus lucidus Turcz and irbesartan for 2 days,and then the expression of glycocalyx core proteins Syndecan-1,Glypican-1 and CD31,α-SMA in rat GECs were detected via Western blot,the level of mRNA was detected by RT-PCR.Results:The expression of glycocalyx core proteins Syndecan-1,Glypican-1,CD31 and their mRNA decreased,while the expression ofα-SMA and its mRNA increased in rat GECs cultured in high glucose environment.The expression of Syndecan-1,Glypican-1,CD31 and their mRNA was up-regulated andα-SMA and its mRNA expression was inhibited by Lycopus lucidus Turcz and irbesartan-containing serum.The protective effects of Lycopus lucidus Turcz on Syndecan-1 and Glypican-1 was better than that of irbesartan in a dose-dependent manner,and the inhibitory effect of irbesartan onα-SMA was more significant.Conclusion:Lycopus lucidus Turcz attenuates GECs damage induced by high glucose by protecting glycocalyx core proteins,which provides another important theoretical basis for the treatment with DN.
