摘 要:目的研究单叶蔓荆提取物对小鼠的抗炎作用及其初步作用机制。方法小鼠60只,随机分为阴性对照组(生理盐水)、阳性对照组(阿司匹林,0.4 g/kg)以及单叶蔓荆提取物组低、中、高剂量组(按照生药计分别为10,30,90 g/kg灌胃给药)。连续灌胃给药7 d后,采用小鼠耳肿胀法观察其抗炎作用;另取小鼠60只,同样分组给药后,采用小鼠足肿胀法观察其抗炎作用;ELISA法测定血浆中炎症相关细胞因子TNF-α,IL-1β,IL-6以及PGE_(2)水平;并同时测定肿胀足趾中的MDA以及NO含量。结果与空白组相比,阳性对照组和单叶蔓荆提取物低、中、高剂量组均可显著性降低二甲苯所致小鼠耳肿胀程度(P<0.05或者P<0.01),抑制率分别为31.5%,56.5%和80.4%;阳性对照组和单叶蔓荆不同剂量组小鼠在角叉菜胶致炎后0.5 h、1 h、2 h和4 h的足肿胀率显著降低(P<0.01);阳性对照组(给药阿司匹林)单叶蔓荆中剂量和高剂量提取物组中的炎症因子TNF-α,IL-1β,IL-6以及PGE_(2)含量均呈显著性降低(P<0.05或者P<0.01);阳性对照组(给药阿司匹林)单叶蔓荆中剂量和高剂量提取物组均可引起炎症小鼠足趾组织中MDA和NO含量显著性降低(P<0.01)。结论单叶蔓荆提取物对二甲苯所致耳肿胀、角叉菜胶所致足肿胀模型小鼠均有显著炎症抑制作用,其作用机制与降低体内TNF-α、IL-1β和IL-6等炎症细胞因子表达,抑制体内PGE_(2)合成、减少炎性组织中NO和MDA释放有关。Objective The research aimed to study the anti-inflammatory effect of the extract of Vitex trifolia L.(VT),and to explore the related mechanism.Methods Sixty mice was randomly divided into five groups,blank control group(normal saline),positive control(aspirin),VT extract low-dose,medium-dose and high dose groups(10,30,90 g/kg,by crude medicine).Ear swelling of mice was induced by xylene after intragastric administration for consecutive 7 days.And another sixty mice were grouped and treated by intragastric administration the same way,following by carrageenan-induced swelling of mouse foot plantar.The contents of TNF-α,IL-1β,IL-6 and PGE_(2)in the mice plasma were determined by ELISA methods.The content of NO and MDA in the plantar inflammatory tissue induced by carrageenan of the mice were determined by nitrate reductase method.Results Compared with the blank control group,aspirin group and each group of VT extract could significantly inhibit the swelling degree of mice(P<0.05 or P<0.01)with the inhibitory rates of 31.5%,56.5%and 80.4%,and also the paw swelling rates of mice in aspirin group and all of the VT extract group were decreased significantly.The contents of TNF-α,IL-1β,IL-6 and PGE_(2)in the mice plasma of the aspirin group,medium-dose and high dose groups were decreased significantly,after inducing inflammation.The contents of NO and MDA in the plantar inflammatory tissue induced by carrageenan of the mice of the aspirin group,medium-dose and high dose groups were also decreased significantly.Conclusion The extract of Vitex trifolia L.showed obvious anti-inflammatory effects on ear swelling induced by xylene and paw swelling inducing by carrageenan,which might be related with its ability of inhibiting the expression of TNF-α,IL-1β,IL-6 and PGE_(2),the reduction of NO and MDA release.
