摘  要:目的:观察通管消癥饮(TGXZ)对输卵管炎性不孕症大鼠的影响,探讨其可能的分子机制。方法:选取6~8周龄SD雌性大鼠92只,采用混合菌阴道接种法构建输卵管炎性不孕症大鼠模型。将造模成功的60只大鼠随机分为模型组、西药组(甲硝唑片和盐酸左氧氟沙星胶囊)、低剂量TGXZ组(5.33 g·kg;)、中剂量TGXZ组(10.67 g·kg;)和高剂量TGXZ组(21.33 g·kg;),每组12只,另取12只正常大鼠作为正常对照组。所有实验动物每日灌胃2次,早晚各1次,共1剂,共灌胃30 d。观察并记录大鼠精神状态、活动量、饮水量和爪色等情况,采用ELISΑ法测定各组大鼠血清中白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、白细胞介素10(IL-10)和肿瘤坏死因子α(TNF-α)水平,苏木精-伊红(HE)染色法观察各组大鼠输卵管组织病理形态表现,蛋白免疫印迹(Western blotting)法检测各组大鼠输卵管组织中Toll样受体4(TLR4)/核因子κB(NF-κB)信号通路相关蛋白表达水平;观察各组大鼠受孕率。结果:正常对照组大鼠精神状态良好,活动量和饮水量正常,爪色粉白;模型组大鼠精神状态萎靡,活动量和饮水量降低,爪色暗红;中剂量TGXZ组、高剂量TGXZ组和西药组大鼠的一般情况得到明显改善,大鼠精神状态恢复,活动量和饮水量增加,爪色颜色变浅。与正常对照组比较,模型组大鼠血清炎症因子IL-1β、IL-6、IL-10和TNF-α水平明显升高(P<0.05),输卵管组织炎性细胞浸润评分明显升高(P<0.05),输卵管组织中TLR4、磷酸化NF-κB抑制剂α(p-IκBα)和NF-κB p65蛋白表达水平明显升高(P<0.05),IκBα蛋白表达水平明显降低(P<0.05),大鼠受孕率明显降低(P<0.05);与模型组比较,中和高剂量TGXZ组及西药组大鼠血清中IL-1β、IL-6、IL-10和TNF-α水平明显降低(P<0.05),输卵管组织炎性细胞浸润评分明显降低(P<0.05),输卵管组织中TLR4、p-IκBα和NF-κB p65表达水平均明显降低(P<0.05),IκBα表达水平明显�Objective:To investigate the effect of Tongguan Xiaozheng Decoction(TGXZ)in the rats with tubal inflammatory infertility,and to explore its possible molecular mechanism.Methods:A total of92 SD female rats aged 6-8 weeks were selected,and the rat models of tubal inflammatory infertility were constructed by vaginal inoculation with mixed bacteria.The 60 model rats were randomly divided into model group,western medicine(metronidazole tablets and levofloxacin hydrochloride capsules)group and low(5.33 g·kg^(-1)),medium(10.67 g·kg^(-1)),and high(21.33 g·kg^(-1))doses of TGXZ groups,with12 rats in each group,and another 12 normal rats were taken as normal control group.All rats were intragastrically administered twice a day,once in the morning and evening,for a total of 1 dose,for 30 d.The mental state,activity levels,water consumptions,paw color,etc.of the rats in various groups were observed and recorded;ELISA method was used to measure the levels of serum interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-10(IL-10),tumor necrosis factor-α(TNF-α)of the rats in various groups;Hematoxylin eosin(HE)staining was used to observe the pathomorphology of fallopian tube tissue of the rats in various groups;Western blotting method was used to detect the expression levels of Toll-like receptor 4/nuclear factor-kB(TLR4/NF-κB)signaling pathway related proteins in the fallopian tube tissue of the rats in various groups;the conception rates of rats in various groups were calculated.Results:The rats in normal control group had good mental state,normal activity and drinking water,and white claw color.The mental state of the rats in model group was depressed,the amount of activity and drinking water were decreased,and the claw color was dark red.The general situation of rats in medium dose of TGXZ group,high dose of TGXZ group and Western medicine group was significantly improved,the mental state of rats recovered,the amounts of activity and drinking water were increased,and the color of claw became lighter.Compared with nor
