摘 要:【目的】通过开发和筛选与核桃黑斑病抗性相关的SSR分子标记实现对不同核桃品种黑斑病高效准确的抗性鉴定。【方法】基于核桃转录组测序(RNA-Seq)结果,开发出可能与抗性相关的SSR标记,在抗感差异材料中验证与核桃黑斑病抗性相关的SSR引物。【结果】以12份抗病株系和感病株系基因组DNA为模板,获得1个与核桃抗黑斑病相关的SSR标记,该标记在抗病材料中能够扩增出1条320 bp左右大小的条带(命名为RJR18-M320)。使用此引物对现有25份核桃种质资源进行抗性鉴定,获得抗病材料16份,感病材料9份。【结论】筛选出了1对可以用于核桃抗黑斑病辅助选择的分子标记,加速了核桃抗黑斑病育种进程。【Objective】By developing and screening SSR molecular markers related to walnut bacterial blight’s resistance,this study aims to achieve an efficient and accurate resistance identification of differ⁃ent walnut bacterial blights.【Method】Based on the results of walnut transcriptome sequencing(RNA-Seq),SSR markers that may be related to resistance were developed,and SSR primers related to walnut bacterial blight’s resistance were verified in different anti-inductive materials.【Result】Using the ge⁃nomic DNAs of 12 disease-resistant and infected lines as the template,a SSR marker related to walnut bacterial blight’s resistance was obtained,and this marker could be amplified in the disease-resistant material a band with the length of 320 bp(named RJR18-M320).Using this primer to carry out the resis⁃tance identification of 25 existing walnut germplasm resources,this study obtained 16 resistant materials and 9 susceptible materials.【Conclusion】A pair of molecular markers that can be used for auxiliary se⁃lection of walnut resistance to bacterial blight were screened out,which effectively accelerated the breed⁃ing process of walnuts resistant to bacterial blight.
