摘  要:目的建立南、北五味子直接进样紫外(FI-UV)指纹图谱,用于五味子类药材快速鉴别。方法利用高效液相色谱(HPLC)技术建立南、北五味子FI-UV指纹图谱。对总紫外吸收图谱和紫外光谱进行主成分分析(PCA)和偏最小二乘法分析(PLS-DA)模式分析。结果南、北五味子药材总紫外吸收图谱和紫外吸收光谱轮廓差异明显,呈现不同指纹特性。PCA分析可将两者区分,PLS-DA分析显示紫外光谱中变量211、240和253 nm对其分类起主要作用,进一步分析提示变量240、253 nm对其分类起决定作用。PLS-DA模型R^(2)X、R^(2)Y、Q^(2)均大于0.96,说明模型稳定好,预测能力强。结论FI-UV指纹图谱结合化学计量学方法能够对南、北五味子药材进行快速鉴别及内在质量评价。Objective To establish a fingerprint of Flow-injection Ultraviolet spectroscope(FI-UV)for rapid identification of Schisandra chinensis(Turcz.)Baill.and Schisandra sphenanthera Rehd.et Wils.Methods FI-UV fingerprints of SCB and SSW are established by HPLC.Total absorbance chromatograms and ultraviolet spectra are obtained and analyzed with the aid of principal component analysis(PCA)and partial least-squares discrimination(PLS-DA).Results The total absorbance chromatograms and ultraviolet spectra are different obviously,and displayed fingerprint characteristics.PCA and PLS-DA are successfully used to differentiation of SCB and SSW.The variables of 211,240,253 nm are played important roles in the differentiation of samples,which reflected from the results of PLS-DA.Further analysis indicated that the variables of 240,253 nm played determinative roles.The R^(2)X,R^(2)Y and Q^(2) value of PLS-DA are all above 0.96,which shows the model is stable and have strong predicitive ability.Conclusion FI-UV fingerprint combined with chemometric method could be used for differentiation and quality assessment of SCB and SSW herb source.
