摘  要:以水作为溶剂,在常压与减压下提取苦参碱与氧化苦参碱,对比其提取效果。以乙腈-无水乙醇-3%磷酸(体积比80∶10∶10)作为流动相,检测波长220 nm的条件下,采用高效液相色谱法,测定提取液中苦参碱与氧化苦参碱的含量。结果显示,常压法最佳提取条件为水料比8∶1,提取次数3次,每次提取时间1 h。在该工艺条件下,测得苦参总碱含量(苦参碱与氧化苦参碱的总量)、苦参碱与氧化苦参碱质量分数分别为1.19%、0.83%和0.36%。减压法最佳提取工艺的水料比、提取次数、提取时间与常压法相同,但是提取温度为70℃,最终检测苦参总碱以及各自的含量分别为1.20%、0.08%和1.12%。结果表明,两种提取工艺的苦参总碱的总质量值区别不大,但是减压法提取的氧化苦参碱含量更高,因此减压提取工艺能够有效减少毒性成分苦参碱的比例,降低苦参类药物的总体毒性,增强其推广应用价值。The extraction effects of matrine and oxymatrine under atmospheric and reduced pressures in water as solvent was compared.The contents of matrine and oxymatrine in extracts were measured by HPLC.The mobile phase consisted of acetonitrile,anhydrous ethanol and 3%phosphoric acid(volume ratio was 80∶10∶10),and the detection wavelength was 220 nm.The results showed that the optimum extraction conditions of atmospheric method were as follows:the ratio of water and extraction material was 8∶1,the extraction frequency was 3 times,and duration of extraction in each time was 1 h.The total content and the single contents of matrine and oxymatrine were 1.19%,0.83%and 0.36%,respectively under extraction conditions described above.As for the optimized process of reduced pressures method,the ratio of water and extraction material,extraction frequency and duration were same as that of atmospheric method,however,the extraction temperature was 70℃,and optimum detection results showed that the total content and single contents of matrine and oxymatrine were 1.20%,0.08%and 1.12%.The results described above indicated that under the two extraction processes,the total mass values of matrine and oxymatrine had no significant difference.Nevertheless,the content of oxymatrine in reduced pressures was significantly higher than that in atmospheric pressure,which means that reduced pressures extraction can effectively decrease the ratio of toxic matrine,reduce the general toxicity of Sophora flavescens contained drugs,and enhance their application value.
