摘 要:目的:观察支气管哮喘小鼠的气道重塑状态,探讨中药补肺颗粒对气道重塑的影响及作用机制。方法:以OVA·Al(OH)_(3)混合液腹腔注射致敏联合雾化吸入OVA激发的方法制作哮喘小鼠模型,随机分为四组:正常对照组、哮喘模型组、补肺颗粒组、地塞米松组,分别给予相应药物灌胃干预,末次激发24 h内处死取材。取左肺肺泡灌洗液(BALF)行ELISA法检测转化生长因子β_(1)(TGF-β_(1))、肿瘤坏死因子α(TNF-α)、血管内皮生长因子(VEGF)、内皮抑素(ES)等的浓度。右肺上中叶组织行HE染色、Masson染色、AB-PAS染色,右肺下叶组织以RT-PCR法检测TGF-β_(1)、TNF-α、VEGF、ES等的mRNA表达。结果:同哮喘模型组相比,补肺颗粒组及地塞米松组气管壁及管周炎症细胞浸润、黏膜增厚减轻,胶原沉积减少,气道上皮内的AB-PAS染色阳性信号区域减少。同哮喘模型组对比,补肺颗粒组小鼠肺组织中TNF-α、TGF-β_(1)的mRNA水平有下降趋势,BALF中的TNF-α、TGF-β_(1)水平均有降低,其中TNF-α差异有统计学意义(P<0.05)。补肺颗粒组小鼠肺组织中VEGF、ES的mRNA水平均有降低,差异有统计学意义(P<0.05),小鼠BALF中VEGF、ES的水平均有降低,其中VEGF差异有统计学意义(P<0.05)。结论:补肺颗粒能有效抑制哮喘小鼠气道重塑,其作用机制可能与干预TGF-β_(1)/TNF-α作用的成纤维信号通路以及调节气道血管生成有关。Objective:To observe the status of airway remodeling in asthmatic mice and explore the effect and mechanism of Bufei granule on airway remodeling.Methods:Asthmatic mice model was prepared by intraperitoneal injection of OVA·Al(OH)_(3) mixture combined with OVA inhalation.Mice model were randomly divided into 4 groups:normal control group,asthma model group,Bufei granule group,dexamethasone group,which were separately given with the corresponding drug and then killed after the last stimulation.The pathological changes of lung and trachea were observed by HE staining,Masson staining and AB-PAS staining,and the levels of TGF-β_(1),TNF-α,VEGF and ES in lung tissue and BALF were detected.Results:Compared with asthma model group,the infiltration of inflammatory cells,mucosal thickening,collagen deposition,AB-PAS staining positive area in Bufei granule group and dexamethasone group were reduced.Compared with asthma model group,the mRNA levels of TNF-αand TGF-β_(1) in lung tissue of mice in Bufei granule group showed a downward trend,while the levels of TNF-αand TGF-β_(1) in BALF were decreased(P<0.05).The mRNA levels of VEGF and ES in the lung tissue of mice in Bufei granule group were decreased(P<0.05),while the levels of VEGF and ES in BALF of mice were decreased,either(P<0.05).Conclusion:Bufei granule can effectively inhibit airway remodeling in asthmatic mice,and its mechanism may be related to the intervention of TGF-β_(1)/TNF-αfibroblast signaling pathway and the inhibition of airway angiogenesis.
