摘  要:以枸杞花药离体培养获得的植株根尖为材料,采用植物染色体制片的方法,研究剪根时间、取样时间、预处理时间及解离时间对染色体制片效果的影响,以探寻适宜枸杞花培苗根尖染色体的制片技术和倍性鉴定技术。结果表明:取样时根系长度为1.0~1.5 cm,取样时间在10:00~12:00,于4℃温度条件下,在0.2%的秋水仙素溶液中预处理7~8 h,常温下采用酸解法解离8~10 min后用改良石碳酸复红染液进行涂片滴染所获得的枸杞根尖染色体制片效果最好。Using the root tips of anther culture seedlings which were obtained from wolfberry as materials,the method of chromosome preparation was used,the time of cutting,sampling,pretreatment,dissociation were studied to explore the suitable methods for the technique of chromosomes preparation and ploidy identification.The results showed that Chromosome preparation is the best,when the root grows to 1.0~1.5 cm,take the root tips from 10:00to 12:00,which are pretreated with 0.2% colchicine solution for 7~8 h at temperature of 4 ℃,and dissociated for8~10 min at room temperature,and dripped with modified stone carbonic acid red dyeing solution.
