摘 要:为研究杨树根系响应铵态氮胁迫的机理,以杨树“南林3412杨”无菌苗为试验材料,用NH;Cl溶液胁迫处理后,利用iTRAQ标记定量蛋白质组学技术,分析不同质量浓度的铵态氮胁迫处理下杨树根系的差异表达蛋白。结果表明:一共鉴定到266个差异蛋白,这些差异蛋白按KEGG分类,主要富集在能量代谢、氨基酸代谢和次生代谢物质合成等代谢途径。进一步分析发现,包括果胶酶、木质素过氧化物酶和角质酶等在内的11个细胞壁相关的蛋白在铵态氮胁迫下发生了显著的差异表达,参与糖酵解代谢、碳水化合物代谢的蛋白更倾向于在高质量浓度铵态氮胁迫下上调表达。此外,杨树中有17个抗氧化系统酶和5个热激蛋白在铵态氮胁迫下差异表达。上述结果表明,杨树根系主要通过维持细胞壁完整性、调节碳和能量代谢、激活抗氧化系统等方式来应对铵态氮胁迫。In order to investigate the mechanisms of poplar roots in response to ammonia nitrogen stress, Populus deltiodes cv. ‘3412’ was treated with NH4 Cl solution for five weeks. After that, iTRAQ labeling quantitative proteomics was used to analyze the differentially expressed proteins in poplar roots under different concentrations of ammonia nitrogen stress. A total of 266 differential proteins were identified. According to KEGG pathway classification, these differential proteins were mainly concentrated in energy metabolism, amino acid metabolism, and secondary metabolite synthesis. Further, 11 cell wall related proteins including pectinase, lignin peroxidase, and cutinase, were differentially expressed. Proteins involved in glycolysis and carbohydrate metabolism were more likely to be up-regulated under high ammonia nitrogen stress. In addition, 17 antioxidant system enzymes and 5 heat shock proteins were differentially expressed in poplar under ammonia nitrogen stress. Under ammonia nitrogen stress, a variety of proteins in poplar roots participate in ammonia nitrogen responses. They mainly use the ways of maintaining cell wall integrity, regulating carbon and energy metabolism, activating antioxidant system and other ways to resist ammonia nitrogen stress.
