摘  要:选取鸢尾叶片为试验材料,通过3种不同辐射强度(分别为168、210、252μw/cm^(2))的UV-B辐射,在不同辐射时间(第3、7、14、21 d)取样测试,根据关键酶活性计算公式处理实验结果,采用Excel绘制出不同辐射强度梯度和时间梯度下横向与纵向对比的酶含量变化曲线,探究辐射强度及辐射处理时间对酶活性的响应机制,考察了紫外可见分光光度法探究UV-B辐射增强对鸢尾叶片中APX、PAL、GR关键酶活性的影响。结果表明:鸢尾中不同的关键酶抗辐射能力不同,在辐射处理时间不变且辐射强度为252μw/cm^(2)时,PAL、APX酶活性较好,抗辐射能力强,GR酶在辐射强度为168μw/cm^(2)时,活性较好,不能承受高强度辐射;当辐射强度不变且辐射处理时间在第7 d时,PAL、GR酶含量较高,APX酶在辐射时间第14 d时,含量较高,酶活性较好,鸢尾叶片中APX、PAL、GR的活性对UV-B辐射变化响应的敏感性存在差异。In this study,Iris leaves are selected as the test material.By three different radiation intensity(respectively 168μw/cm^(2),210μw/cm^(2),252μw/cm^(2))of UV-B radiation,in different radiation time(day 3,day 7,day 14,day 21)sampling test.The experimental results are processed according to the key enzyme activity calculation equation,the graphs of the changes in enzyme content for different radiation intensity gradients and time gradients are plotted horizontally versus vertically using Excel.To investigate the response mechanism of radiation intensity and radiation treatment time on enzyme activity,the Ultraviolet visible spectrophotometric method is investigated to probe the effect of UV-B radiation enhancement on the key enzyme activities of APX,PAL and GR in Iris leaves.The results show that different key enzymes in Iris are differently resistant to radiation,PAL and APX enzymes have better activity and are more resistant to radiation when the radiation treatment time is constant and the radiation intensity is 252μw/cm^(2),GR enzymes have better activity when the radiation intensity is 168μw/cm^(2) and cannot withstand high intensity radiation,PAL and GR enzymes are higher when the radiation intensity is constant and the radiation treatment time is at day 7,and APX enzymes are higher and have better enzyme activity at day 14 of the radiation time.This suggests that there are differences in the sensitivity of the activities of APX,PAL,and GR in Iris leaves in response to changes in UV-B radiation.
