摘  要:目的优化洋葱醇提物中总皂苷含量测定的显色条件,并评价抗肿瘤活性。方法采用单因素试验优选洋葱醇提物中总皂苷的显色条件,采用香草醛-硫酸比色法测定其含量,采用CCK-8试剂盒测定其对HepG2细胞、HeLa细胞、A549细胞的抑制作用。结果洋葱醇提物中总皂苷的最佳显色条件为使用5%香草醛-冰醋酸0.4 mL,80%硫酸0.6 mL,75℃下反应15 min,冰浴20 min;洋葱醇提物作用24 h后对HepG2细胞、HeLa细胞、A549细胞的半数抑制浓度(IC50)分别为4.463,7.126,3.964 mg/mL,作用48 h后对上述3种细胞的IC_(50)分别为3.371,5.551,8.988 mg/mL。结论该显色条件可客观评价洋葱醇提物中总皂苷的含量,且洋葱醇提物具有抑制HepG2细胞、Hela细胞、A549细胞生长的作用。Objective To optimize the chromogenic conditions for the determination of total saponins in the ethanol extract from Allium cepa,and to evaluate its antitumor activity.Methods The chromogenic conditions of total saponins in the ethanol extract from Allium cepa were optimized by the single-factor test,the content of total saponins in the ethanol extract from Allium cepa was determined by vanillin-sulfuric acid colorimetry method,and the inhibitory effects of the ethanol extract from Allium cepa on the HepG2 cells,HeLa cells and A549 cells were determined by cell counting kit-8(CCK-8).Results The optimal chromogenic conditions of total saponins in the ethanol extract from Allium cepa were as follows:using 0.4 mL of 5%vanillin-glacial acetic acid and 0.6 mL of 80%sulfuric acid,reaction at 75℃for 15 min and ice bath for 20 min.The half-maximal inhibitory concentration(IC50)of the ethanol extract from Allium cepa on the HepG2 cells,HeLa cells and A549 cells after 24 h was 4.463,7.126,3.964 mg/mL,the IC50 of the ethanol extract from Allium cepa on the above three kinds of cells after 48 h was 3.371,5.551,8.988 mg/mL.Conclusion The chromogenic conditions can objectively evaluate the content of total saponins in the ethanol extract from Allium cepa,and the ethanol extract from Allium cepa can inhibit the growth of HepG2 cells,Hela cells and A549 cells.
