摘  要:目的为肝细胞癌(HCC)的靶向药物治疗提供新思路。方法检索TCGA数据库中的HCC RNA芯片数据集,比较数据集中AGTR1与血管内皮生长因子A(VEGFA)的表达是否具有相关性;通过体内外实验研究AGTR1过表达对HCC细胞生长的影响。结果AGTR1与VEGFA的表达具有相关性,且随着AGTR1表达的上升相关性增强。通过对HCC细胞株BEL-7404的体内外实验发现,AGTR1的过表达能引起VEGFA的表达上调,进而促进HCC细胞的生长;洛沙坦对过表达AGTR1的BEL-7404细胞具有一定抑制作用,可抑制VEGFA蛋白的表达。结论AGTR1抑制剂对AGTR1过表达引起的VEGFA上调具有抑制作用,为进一步研究AGTR1抑制剂应用于AGTR1高表达HCC患者的靶向治疗提供了参考。Objective To provide new ideas for targeted drug therapy of hepatocellular carcinoma(HCC).Methods The RNA microarray dataset of HCC patients in the TCGA database was searched to compare whether the expression of AGTR1 and vascular endothelial growth factor A(VEGFA)in the dataset was correlated.The effect of AGTR1 overexpression on the growth of HCC cells was studied by in vivo and in vitro experiments.Results The expression of AGTR1 and VEGFA was correlated,and the correlation was increased with the increase of AGTR1 expression.The in vitro and in vivo experiments on the HCC cell line BEL-7404,showed that AGTR1 overexpression could increase the expression of VEGFA and promote the growth of HCC cells.Losartan had a certain inhibitory effect on BEL-7404 cells overexpressing AGTR1,and could inhibite VEGFA protein expression.Conclusion AGTR1 inhibitor has a inhibitory effect on the up-regulation of VEGFA caused by AGTR1 overexpression,which provides a reference for the further study of AGTR1 inhibitor in the targeted therapy of HCC patients with high AGTR1 expression.
