摘 要:[目的]通过试验考察生品及醋炙品夏天无水提取物的急性毒性以及镇痛作用。[方法]采用Bliss法计算生品、醋炙夏天无水提取物的半数致死量(LD_(50)),连续观察14 d后进行解剖学检查,计算小鼠心、肝、脾、肺、肾脏器指数,检测血液生化指标(ALT、AST、CK、LDH、UREA、CREA)的水平,观察各脏器HE染色病理切片结果;以冰醋酸致小鼠扭体反应对夏天无及其醋炙品提取物进行镇痛评价。[结果]生品夏天无和醋炙夏天无的原生药半数致死量(LD_(50))分别为48、54 g/kg;急性毒性差异不明显,分别相当于人体(体质量60 kg)安全用量的240和270倍;与正常组相比,生化指标除AST有差异外(P<0.05),ALT、CK、LDH、UREA、CREA的水平差异不显著(P<0.05);与正常组对比,醋炙夏天无组能明显减少小鼠扭体次数(P<0.05)。[结论]夏天无及其醋炙品水提取物LD_(50)>10 g/kg原生药,属于实际无毒性,生品夏天无的毒性作用略大于醋炙品夏天无,说明醋炙夏天无具有一定的减毒作用;醋炙后镇痛作用增强,进一步阐明醋炙入肝、增强止痛这一理论。[Objective]To investigate the acute toxicity and analgesic effect of the Corydalis decumbens anhydrous extract of raw products and vinegar products through experiments. [Method]The Bliss method was used to calculate the LD_(50)of raw and the stir-frying with vinegar of Corydalis decumbens extracts. After 14 days of continuous observation, anatomical examination was performed to calculate the index of heart, liver, spleen, lung, and kidney organs in mice, detect the levels of blood biochemical indexes ALT, AST, CK, LDH, UREA, and CREA,and observe the HE staining pathology of each organ.Analgesic evaluation of Corydalis decumbens and its vinegar-cured extracts by glacial acetic acid-induced writhing reaction in mice.[Result]The crude lethal doses(LD_(50)) of the raw medicine Corydalis decumbens and its vinegar products were 48 and 54 g/kg, respectively;the acute toxicity difference was not obvious, which was equivalent to 240 and 270 of the safe dosage of human body(body weight 60 kg), respectively.Compared with the normal group, except for AST, there were differences in biochemical indexes(P<0.05), but the levels of ALT, CK, LDH, UREA and CREA were not significantly different(P>0.05).Compared with the normal group, the stir-frying with vinegar of Corydalis decumbens group could significantly reduce the number of writhing in mice(P<0.05). [Conclusion]The LD_(50)>10 g/kg native medicine of Corydalis decumbens and the stir-frying with vinegar of Corydalis decumbens extracts is actually non-toxic. The toxic effect of raw Corydalis decumbens is slightly greater than the stir-frying with vinegar of Corydalis decumbens,indicating that the stir-frying with vinegar of Corydalis decumbens has a slight detoxification effect after sunburn.The analgesic effect is enhanced after the processing of the stir-frying with vinegar, further elucidating the theory that the processing of the stir-frying with vinegar enters the liver and enhances the analgesic effect.
