摘 要:目的研究参附注射液对幼鼠脓毒血症急性肝损伤的保护机制。方法48只幼鼠随机分为空白对照组(Ctrl)、脓毒血症组(LPS)、治疗组(SFI),每组16只。幼鼠脓毒血症模型构建:腹腔注射脂多糖10 mg/kg。参附注射液治疗组构建:于60 min后腹腔注射参附注射液10 mL/kg。分别于6 h、12 h检测血清ALT,AST、肝组织中的TNF-α、肝组织乙酰胆碱、肝组织乙酰胆碱转移酶以及两者同TNF-α的联系。结果与LPS组比较:SFI组在6 h、12 h血清ALT、AST、肝组织中的TNF-α都显著降低(P<0.01),肝组织乙酰胆碱、乙酰胆碱转移酶都显著升高(P<0.01)。LPS组的肝组织中的TNF-α与乙酰胆碱、乙酰胆碱转移酶都呈负相关(P<0.01)。结论参附注射液影响胆碱能抗炎通路机制保护脓毒血症急性肝损伤。Objective To study the protection of Shenfu Injection(SFI)on septic acute liver injury in young mice.Methods 48 young mice were divided at random into three groups:the blank control group(Ctrl,n=16),the sepsis group(LPS,n=16),the therapeutic in Shenfu Injection group(SFI,n=16).The septic model on young mice was established by intra-peritoneal injection of lipopolysaccharide(LPS)at 10 mg/kg.Shenfu Injection Group:At 60 minutes the septic young mice were injected Shenfu Injection(10 mL/kg)on intra-peritoneal.At 6h,12h,ALT and AST in serum,TNF-α,Ach and ChAT in liver tissues were detected,and the correlation between TNF-αwith Ach and TNF-αwith ChAT were analyzed.Results Compare to LPS group,at 6h,12h in the SFI group the serum levels of ALT and AST,the expression of TNF-αin liver tissue were significantly decreased(P<0.01),the content of Ach and the activities of ChAT in liver tissue were significantly increased(P<0.01).In the LPS group(P<0.01)the TNF-αwas negatively correlated with the Ach and ChAT.Conclusion Shenfu Injection affects cholinergic anti-inflammatory pathway to protect acute liver injury of septic young mice.
