摘 要:本文以冬青卫矛基因组DNA为模板,克隆LTR反转录转座子Ty1-copia类与Ty3-gypsy类反转录酶(reverse transcriptase,RT)序列,通过测序及相关生物信息学软件对所获序列变化特点进行分析。最终获得Ty1-copia与Ty3-gypsy类反转录转座子RT序列分别为40条和39条。其中Ty1-copia-RT序列长度在231~267 bp之间,核酸序列的相似性为8.05%~97.7%;Ty3-gypsy-RT序列长度范围为365~499 bp,核酸序列的相似性为10.04%~99.31%。将所获得的核酸序列翻译为氨基酸后发现RT氨基酸序列具有高度异质性。通过与其他物种RT序列进行比对,结果显示冬青卫矛与其他一些植物间的部分RT序列相似性较高,可能存在共同起源,印证了LTR反转录转座子不同物种间可能存在横向传递。In this paper,reverse transcriptase(RT) sequences of Ty1-copia and Ty3-gypsy retrotransposons were obtained through PCR amplification using Euonymus japonicus genomic DNA as template.The obtained sequences were analyzed by sequencing and bioinformatics software.Results revealed that 40 Ty1-copia like RT sequences and 39 Ty3-gypsy like RT sequences were identified.The length of Ty1-copia-RT sequences ranged from 231 bp to 267 bp,and the similarity of nucleic acid sequences ranged from 8.05% to 97.7%.The length of Ty3-gypsy-RT sequences was from 365 bp to 499 bp,and the similarity of nucleic acid sequences ranged from 10.04% to 99.31%.The translated RT amino acid sequences showed high heterogeneity.The phylogenetic analysis of RT sequences showed diverse similarity between E.japonicus and other species,reflecting that they had a common origin,and a horizontal transmission of LTR retrotransposons occurred in the process of evolution among different species.
