摘  要:目的探讨高亲和力IgG受体Fc-gammaⅠ型(FcγRⅠ)在阿尔茨海默病(AD)脑皮层的表达变化。方法选用正常人脑组织样本和神经病理诊断为AD患者脑组织样本,6月龄野生型小鼠和APP/PS1小鼠。免疫荧光和免疫蛋白印迹检测脑皮层FcγRⅠ的细胞定位和表达。结果人和小鼠脑皮层的神经元均普遍表达FcγRⅠ。与健康人脑相比,AD患者脑皮层的FcγRⅠ表达显著降低(P<0.05)。与野生型小鼠脑相比,AD模型小鼠脑皮层的FcγRⅠ表达也呈现出显著降低(P<0.05)。结论IgG高亲和力受体FcγRⅠ在AD患者脑和AD模型小鼠脑皮层的表达显著下降,可能与AD的神经病理机制相关。Objective To explore expression changes of high affinity IgG receptor Fc-gammaⅠtype(FcγRⅠ)in brain cortex of patients with Alzheimer's disease(AD).Methods Normal human brain tissue samples and brain tissue samples with neuropathological diagnosis of AD,wild type mice and APP/PS1 mice were used.Immunofluorescence and Western blot were used to define cellular localization and expression of FcγRⅠin cerebral cortex.Results FcγRⅠwas generally expressed in neurons of human and mouse cerebral cortex.Compared with normal human brain,FcγRⅠexpression in AD human brain cortex decreased significantly(P<0.05).Compared with wild type mouse brain,FcγRⅠexpression in cerebral cortex of AD mouse models also decreased significantly(P<0.05).Conclusions The expression of IgG high affinity receptor FcγRⅠin AD human brain and cerebral cortex of AD mouse models decreases significantly that suggestes that FcγRⅠis potentially correlated with AD neuropathological pathogenesis.
