摘  要:目的:研究天麻素(GAS)对氧糖剥夺(OGD)激活的BV2细胞Notch通路表达的影响。方法:BV2细胞分成4组,分别为对照组(control)、氧糖剥夺组(OGD)、OGD联合0.17 mmol/L GAS处理组(OGD+GAS-L),和OGD联合0.34 mmol/L GAS处理组(OGD+GAS-H)。免疫荧光双标染色和Western Blot检测Notch通路相关蛋白Notch-1、NICD、RBP-JK和Hes-1的表达变化。结果:与control组相比,OGD组中的Notch-1、NICD、RBP-JK和Hes-1表达明显增高(P<0.05);GAS干预后上述蛋白表达均明显降低(P<0.05);而不同浓度的GAS治疗组之间无明显差异(P>0.05)。结论:GAS能抑制OGD诱导的BV2细胞中Notch通路相关分子的表达。Objective:To study the effect of gastrodin(GAS)on the Notch signaling pathway in microglia induced by oxygen glucose deprivation(OGD).Methods:BV2 cells were divided into four groups:control group(control),oxygen glucose deprivation group(OGD),OGD combined with 0.17 mmol/L GAS treatment group(OGD+GAS-L),and OGD combined with 0.34 mmol/L GAS treatment group(OGD+GAS-H).The expression changes of Notch pathway-related protein(Notch-1,NICD,RBP-JK,and Hes-1)were detected by immunofluorescence staining and Western Blot.Results:The results showed that the expression of Notch-1,NICD,RBP-JK,and Hes-1 in OGD group were significantly higher than that in control group(P<0.05);The protein expressions of Notch-1,NICD,RBP-JK,and Hes-1 were significantly decreased after intervention with GAS(P<0.05).However,there was no significant difference between GAS treatment groups with different concentrations(P>0.05).Conclusion:Gas inhibited the expression of Notch pathway related proteins in BV2 cells induced by OGD.
