摘  要:为了解猪接触传染性胸膜肺炎放线杆菌临床分离菌株与标准菌株蛋白质差异表达情况,采用定量差异蛋白质组学研究策略,以临床分离菌株(HN201616)与标准株(CVCC261)的细菌全蛋白质为研究对象,鉴定差异表达蛋白质及生物信息学分析。结果显示:共鉴定出1373个蛋白质,其中显著差异的蛋白质数为395个[显著性差异蛋白质数目按照差异倍数(FC)≥1.5和FC≤0.67,P≤0.05进行相应的筛选],包括上调蛋白质205个,下调蛋白质190个;鉴定出的与抗生素耐药相关的蛋白质主要有二氢叶酸还原酶、外膜蛋白、麦芽糖蛋白、二氢蝶酸合成酶、谷胱甘肽转移酶、丙酮酸激酶、谷氨酰水解酶、ATP-结合盒家族蛋白等。这些结果将有助于深入探索猪接触传染性胸膜肺炎放线杆菌多重耐药机制,为研发新型抗菌药剂和筛选潜在药物靶点提供思路。In order to understand the differential protein expression between clinical isolates and standard strains of Actinobacillus pleuropneumoniae(A.pleuropneumoniae)exposed pigs,quantitative differential proteomics was used to identify the differentially expressed proteins and bioinformatics analysis using the total bacterial protein of clinical isolate(HN201616)and standard strain(CVCC261)as research subjects.The results showed that a total of 1373 proteins were identified,among which the number of significantly different proteins was 395(the number of significantly different proteins was screened according to Fold change≥1.5 and Fold change≤0.67,and P≤0.05),including 205 up-regulated proteins,and 190 down-regulated proteins.Proteins related to antibiotic resistance were mainly identified as dihydrogolate reductase,outer membrane protein,maltoporin,dihydropteroate synthase,glutathione transferase,pyruvate kinase,gamma-glutamyl hydrolase,ATP-binding cassettes transporters(ABC),etc.The identification of these differentially expressed proteins offers further leads for elucidation on the mechanism of multidrug resistance in A.pleuropneumoniae,and facilitates the development of novel antimicrobial agents and the selection of potential drug targets against A.pleuropneumoniae.
