脂多糖诱导C57BL/6J小鼠急性炎症对听力损失的影响

摘  要:目的采用脂多糖(LPS)腹腔注射的方法诱导C57BL/6J小鼠机体产生急性炎症,观察急性炎症期小鼠听力情况,并分析其相关机制。方法将C57BL/6J小鼠30只随机分为空白组、LPS组及对照组,每组10只,LPS组予以2.5 mg/kg的LPS,对照组输注相等体积的生理盐水,其余生活条件保持一致。注射LPS 3、7、15 d后,通过听性脑干反应(ABR)评估小鼠听力情况。冰冻耳蜗组织切片HE染色观察耳蜗组织形态学变化。ELASA方法观察小鼠体内TNF-α、IL-1β、IL-6的变化。同时观测小鼠饮水、饮食及体质量的变化。结果与对照组比较,LPS组在注射3、7 d后的听力阈值均明显升高,P<0.05,但是注射15 d后两组听力阈值比较,差异无统计学意义,其余频率下阈值变化不明显。注射LPS 3 d后,LPS组的TNFα,IL-6及NF-κB水平明显高于对照组,P<0.05,IL-1β无明显变化。LPS组螺旋神经节细胞有丢失,血管纹部分空泡化,形态异常。但是,LPS不影响小鼠的饮食饮水及体质量。结论LPS可以引发小鼠耳蜗急性炎症,造成听力损失,主要表现为高频听力损失。Objective To induce acute inflammation in C57BL/6J mice by intraperitoneal injection of lipopolysaccharide(LPS),to observe the hearing of mice during acute inflammation,and to analyze its related mechanism.Methods Thirty healthy 2-month-old male C57BL/6J mice were randomly divided into the blank group,the LPS group and the control group,with 10 mice in each group.The mice in LPS group were given 2.5 mg/kg LPS by intraperitoneal injection,while the control group was infused with the same volume of normal saline by the same injection method,and the rest of the living conditions were the same.Auditory brainstem response(ABR)was used to evaluate the hearing of mice after LPS injection at 3,7 and 15 days.Frozen cochlear tissue sections were stained with HE staining to observe the morphological changes of cochlea.ELASA method was used to observe the changes of TNF-α,IL-1βand IL-6 in mice.At the same time,the changes of drinking water,diet and body weight were observed.Results Compared with those in the control group,the hearing thresholds of 16 kHz and 32 kHz in the LPS injection group increased significantly 3 and 7 days after the injection(P<0.05),while there was no significant difference in hearing threshold between the two groups 15 days after the injection,and the threshold changes at other frequencies were not significant.Three days after the injection of LPS,the levels of TNFα,IL-6 and NF-κβin the LPS group were significantly higher than those in control group(P<0.05),while there was no significant change for IL-1β.spiral ganglion cells were lost,part of stria vascularis vacuolated and morphology was abnormal in the LPS group.However,LPS did not affect the diet,water and body weight of mice.Conclusion LPS can induce acute inflammation of mouse cochlea and cause hearing loss,which is mainly characterized by high-frequency hearing loss.
