摘  要:由于病毒因素的影响,导致水稻的产量以及结实质量难以得到保障,为此,提出基因编辑技术在培育抗病毒水稻育种中的应用研究。以CRISPR/Cas9技术为核心,通过获得的BEL突变体的基因序列,敲掉染色体中表现出相关病毒的抗性功能缺失的基因,并借助Cas9基因序列的优化机制,降低源基因组的敏感性,结合单碱基编辑、基因替换和基因插入3种手段对基因进行精确编辑,将抗病毒序列嵌入到水稻的基因组中,测试结果表明,基因编辑可以提高水稻的抗病毒能力,改善水稻的产量以及结实质量。Due to the influence of virus factors,it is difficult to guarantee the yield and seed setting quality of rice.Therefore,the application research of gene editing technology in breeding virus-resistant rice was put forward.With CRISPR/Cas9 technology as the core,through the obtained gene sequence of BEL mutant,the genes showing the lack of resistance to related viruses in chromosomes were knocked out,and the sensitivity of the source genome was reduced by the optimization mechanism of Cas9 gene sequence,and the genes were precisely edited by means of single base editing,gene replacement and gene insertion,and the antiviral sequences were embedded into the rice genome.The test results show that gene editing could improve the anti-virus ability of rice,and improve the yield and seed quality of rice.
