摘  要:目的:探讨穴位埋线对去势骨质疏松大鼠骨密度(BMD)、骨髓间充质干细胞(BMSCs)、碱性磷酸酶(ALP)蛋白及转化生长因子(TGF-β)信号通路的影响,为穴位埋线对绝经后骨质疏松症的调控机制提供研究依据。方法:将48只3月龄健康雌性SD大鼠,SPF级(Ⅱ级),随机分成正常组、模型组、穴位埋线组、非穴位埋线组,每组12只。正常组:不做任何手术;模型组:行双测卵巢摘除术造大鼠骨质疏松症模型;穴位埋线组(埋线组):给予肾俞、京门穴位埋线治疗;非穴位埋线组:给予非腧穴位置埋线治疗,非经非穴点选各穴旁5mm处,处理时间及疗程同穴位埋线组。正常组与模型组不做任何治疗,各组均饲养3个月。穴位埋线组与非穴埋线组每10日治疗1次,3次为1个疗程,疗程间隔3天,共治疗3个月。治疗后采用双能X线骨密度检测腰椎、股骨BMD;采用免疫组织化学染色法检测大鼠BMSCs中骨钙素(OC)和I型原胶原N-端前肽(PINP)的含量;采用蛋白免疫印迹法(Western blot,WB)检测ALP蛋白表达水平;采用RT-qPCR法检测TGF-β的mRNA表达水平。结果:模型组腰椎、股骨BMD、BMSCs、ALP蛋白及TGF-β信号通路表达水平低于正常组(P<0.05),穴位埋线组和非穴位埋线组腰椎、股骨BMD、BMSCs、ALP蛋白及TGF-β信号通路表达水平高于模型组(P<0.05),且穴位埋线组腰椎、股骨BMD、BMSCs、ALP蛋白及TGF-β信号通路表达水平高于非穴位埋线组(P<0.05)。结论:穴位埋线可提高去势SD大鼠BMD、BMSCs、ALP蛋白及TGF-β信号通路表达水平,揭示了穴位埋线治疗绝经后骨质疏松的分子调控机制,从而为穴位埋线防治和延缓绝经后骨质疏松提供了一定的理论依据。Objective:To investigate the effects of acupoint catgut embedding on bone mineral density(BMD),bone marrow mesenchymal stem cells(BMSCs),alkaline phosphatase(ALP)protein and transforming growth factor(TGF-β)signaling pathway in caste-cured osteoporosis rats,so as to improve the regulation mechanism of the effect of acupoint catgut embedding on postmenopausal osteoporosis.Methods:48 3-month-old healthy female SD rats(SPF gradeⅡ)were randomLy divided into normal group,model group,acupoint embedding group and non-acupoint embedding group,with 12rats in each group.The normal group did not undergo any surgery.Model group:The rat model of osteoporosis was established by double measure ovariectomy.Acupoint embedding group(embedding group):Shenshu and Jingmen acupoint embedding treatment;Non-acupoint embedding group:non-acupoint position embedding treatment,non-acupuncture point selection of 5mm beside each acupoint;The treatment time and course of treatment were the same as the acupoint embedding group.Normal group and model group were fed for 3months without any treatment.Acupoint embedding group and non-acupoint embedding group once every ten days,three times a course of treatment,three days interval,a total of treatment for 3months.BMD of lumbar spine and femur were detected by dual-energy Xray after treatment.The contents of osteocalcin(OC)and N-terminal propeptide(PINP)in BMSCs were determined by immunohistochemical staining.ALP protein expression was detected by Western blot(WB).RT-qPCR was used to detect the mRNA expression level of TGF-β.Results:The expression levels of BMD,BMSCs,ALP protein and TGF-βsignaling pathway in lumbar spine,femoral BMD,BMSCs,ALP protein and TGF-βsignaling pathway in the model group were lower than those in the normal group(P<0.05),and those in the acupoint embedding group and non-acupoint embedding group were higher than those in the model group(P<0.05).The expression levels of BMD,BMSCs,ALP protein and TGF-βsignaling pathway in lumbar spine,femur in acupoint embedding group wer
