摘 要:目的采用生物信息学的方法识别肝脏IR的关键基因,为改善2型糖尿病(T2DM)提供新思路。方法从CTD数据库下载IR相关基因,同时从GEO数据库下载T2DM基因表达谱GSE15653,利用GEO2R在线工具筛选出T2DM的差异表达基因(DEGs),取2个数据集的交集基因为肝脏胰岛素抵抗基因(IR-DEGs)。然后用R软件及STRING在线网站对IR-DEGs分别进行功能富集分析和蛋白质互作网络分析,并使用Cytoscape软件筛选关键基因。结果筛选得到1197个IR-DEGs,其中在GSE15653中上调IR-DEGs 352个,下调IR-DEGs 845个。基因本体分析提示IR-DEGs主要富集于类固醇激素反应、Hsp90蛋白结合、热休克蛋白绑定、转录辅助激活、多肽结合等。通路分析提示,IR-DEGs主要富集于磷酸戊糖途径、ErbB信号通路、MAPK信号通路、胰岛素信号通路等。利用Cytoscape筛选出10个关键基因,分别是MAPK3、SRC、MAPK8、CCNB1、DLGAP5、ASPM、KRAS、NRAS、NUSAP1、BIRC5。结论通过生物信息学分析可以发现肝脏胰岛素抵抗在2型糖尿病发展中起的新机制。Objective To investigate the key gene of liver insulin resistance(IR)by bioinformatics in patients with type 2 diabetes mellitus(T2DM),so as to provide new ideas for the improvement of T2DM.Methods IR-related genes were downloaded from CTD database,and T2DM gene expression profile GSE15653 was downloaded from GEO database.Differentially expressed genes(DEGs)of T2DM were screened by GEO2R online tool.The intersection genes of two datasets were identified as hepatic insulin resistance genes(IR-DEGs).Moreover the functional enrichment analysis and protein interaction network analysis were performed for IR-DEGS by R software and STRING online website,respectively.In addition the key genes were screened by Cytoscape software.Results A total of 1197 IR-DEGs were obtained,in which,352 were up-regulated and 845 were down-regulated in GSE15653.Gene objective analysis showed that IR-DEGs were mainly enriched in steroid response,Hsp90 protein binding,heat shock protein binding,transcription-assisted activation and polypeptide binding.The signle pathway analysis indicated that IR-DEGs were mainly enriched in pentose phosphate pathway,ErbB signaling pathway,MAPK signaling pathway and insulin signaling pathway,and 10 key genes were screened by Cytoscape,including MAPK3,SRC,MAPK8,CCNB1,DLGAP5,ASPM,KRAS,NRAS,NUSAP1 and BIRC5.Conclusion Bioinformatics analysis reveals a new mechanism of hepatic insulin resistance in the development of T2DM.
