摘  要:为进一步提高野生动物疫情监测、病原检测鉴定和应急防控的时效性,建立了对鸟类拭子样品直接进行禽流感病毒(Avian influenza virus,AIV)核酸检测鉴定的二次PCR方法。对比试验结果显示,该方法可行性较高,具有与实时荧光定量PCR(real-time quantitative PCR,qPCR)方法等同的特异性和敏感性。禽流感病毒核酸二次PCR检测方法的建立,有利于基层监测单位在缺乏病毒分离技术和qPCR等实验条件的情况下,利用现有条件,就近、及时地开展野鸟样品的禽流感病毒检测工作。In order to further improve the effectiveness of wildlife epidemic monitoring,pathogen detection and emergency prevention and control,this study established a secondary PCR method,which can identify AIV nucleic acids directly from swab samples of wild birds.Comparative experimental results show that this method is highly feasible and has the same specificity and sensitivity compared with qPCR.The establishment of the second PCR detection method of AIV nucleic acid is conducive to substrate monitoring stations to carry out the detection of AIV by using the existing conditions in the absence of experimental conditions and technologies,such as virus isolation and qPCR,so that the sample detection can be completed at the nearest site and in a timely manner.
