摘 要:目的:以含药血清为对照,通过检测分析麻杏石甘汤(Maxing Shigan Decoction, MSD)含药肝孵育液的显著性差异成分及其含量、变异系数,为含药肝孵育液的质量控制提供参考,初步探讨其用于MSD体外药效学研究的可行性。方法:应用三重四级杆液-质联用(TSQ-Quantis)技术,采集13组MSD水煎液数据,采用正交偏最小二乘判别分析法分析得到水煎液的差异性成分;采集不同批次MSD含药肝孵育液和含药血清数据,找到二者的差异性成分,计算各成分的含量并分析其RSD。体外药效学实验分为正常组、模型组、含药肝孵育液(2.5%,5%,10%)和含药血清(2.5%,5%,10%)组,用磷酸组胺处理复制支气管上皮细胞损伤模型,分别给予相应药物孵育36 h后,Western blot检测各组磷脂酰肌醇-3激酶(PI3K)和蛋白激酶B(Akt)蛋白的相对表达量,并计算RSD。结果:与水煎液的8个差异性成分相比,含药肝孵育液和含药血清的差异性成分均有7个;含药肝孵育液中差异性成分的含量高,且不同批次间各成分的RSD均小于10%。含药肝孵育液组和含药血清组(2.5%)PI3K蛋白表达降低(P<0.05),含药肝孵育液和含药血清组Akt蛋白的表达均显著降低(P<0.05),且各含药肝孵育液组PI3K和Akt相对表达量的RSD均小于10%。结论:MSD含药肝孵育液中的差异性成分近似于含药血清,该体系成分含量高、稳定性好;对上皮细胞损伤模型PI3K和Akt蛋白表达的作用趋势与含药血清一致,且结果重现性好;结合其模拟口服中药肠吸收后经肝入血的全过程等优点,初步表明含药肝孵育液用于MSD体外药效学研究有一定的可行性。OBJECTIVE To detect and analyze the significantly differential components, content and coefficient of variation of Maxing Shigan Decoction(MSD) medicated liver incubation solution with medicated serum as a control to provide reference for the quality control of medicated liver incubation solution, and to preliminarily discuss the application feasibility in the in vitro pharmacodynamics research of MSD.METHODS Totally 13 groups of MSD data were collected by using triple quadrupole liquid-mass spectrometry(TSQ-Quantis) technology, and the differential components were searched by using orthogonal partial least squares discriminant analysis;the data of different batches of MSD medicated liver incubation solution and medicated serum were collected, and the differential components of the two were explored. The content of each component was calculated and the RSD value was analyzed. The in vitro pharmacodynamic experiment was included control group, model group, medicated liver incubation solution(2.5%, 5%, 10%) groups and medicated serum(2.5%, 5%, 10%) groups. Except the control group, the other groups were treated with histamine phosphate to replicate the bronchial epithelial cell injury model. After 36 hours of incubation with corresponding drug, Western blot was used to detect the relative expression of phosphatidylinositol-3 kinase(PI3 K) and protein kinase B(Akt) proteins in each group, and then the RSD value was calculated.RESULTS Compared with the 8 differential components in the water decoction, there were 7 differential components in both the medicated liver incubation solution and the medicated serum;the contents of 7 significantly differential components in the medicated liver incubation solution were high, and the RSD value of each component among different batches was less than 10%. The medicated liver incubation solution and medicated serum(2.5%) reduced the expression of PI3 K protein(P<0.05), and medicated liver incubation solution and medicated serum(2.5%, 5%, 10%) significantly reduced the expression
